The plant sterol guggulsterone has been shown to exert anti-tumor effects, making it a candidate chemotherapeutic agent. We investigated the anti-tumor effects of guggulsterone on colon cancer cells and elucidated the underlying molecular mechanisms related to angiogenesis. The apoptotic effects of guggulsterone were examined by cell survival assay. Western blot analysis was used to determine the levels of various down-stream intracellular proteins involved in angiogenesis, including signal…

Z-Guggulsterone Induces Apoptosis in Gastric Cancer Cells through the Intrinsic Mitochondria-Dependent Pathway
TLDR
Inhibition of FXR signaling with z-guggulsterone induced anticancer effects in SGC-7901 cells by decreasing cell proliferation and promoting apoptosis through the mitochondria-dependent pathway.
Inhibition of STAT3 signaling and induction of SHP1 mediate antiangiogenic and antitumor activities of ergosterol peroxide in U266 multiple myeloma cells
TLDR
It is suggested that EP can exert antitumor activity in multiple myeloma U266 cells partly with antiangiogenic activity targeting JAK2/STAT3 signaling pathway as a potent cancer preventive agent for treatment of multipleMyeloma cells.
Guggulsterone induces apoptosis of human hepatocellular carcinoma cells through intrinsic mitochondrial pathway.
TLDR
Guggulsterone exerts its anticancer effects by inhibiting cell proliferation and inducing apoptosis in HepG2 cells by activation of the intrinsic mitochondrial pathway.
SR16388: a steroidal antiangiogenic agent with potent inhibitory effect on tumor growth in vivo
TLDR
It is demonstrated that SR16388 at a single daily oral dose of 3 mg/kg for 12 days significantly inhibited the granulation tissue thickness and the microvessel density of the GT as compared to control, and down-regulated the pro-angiogenic transcription factors, hypoxia inducible factor 1α and signal transducer and activator of transcription 3 in non-small cell lung cancer (NSCLC) cells.
Targeting beta-Catenin signaling to induce apoptosis in human breast cancer cells by z-Guggulsterone and Gugulipid extract of Ayurvedic medicine plant Commiphora mukul
TLDR
The present study indicates that the β-Catenin signaling pathway is the target for GL-induced growth inhibition and apoptosis in human breast cancer.
Guggulsterone enhances antitumor activity of gemcitabine in gallbladder cancer cells through suppression of NF-κB
TLDR
Guggulsterone exhibits anticancer activities and enhances the antitumor activities of gemcitabine through the suppression of NF-κB activation in gallbladder cancer cells.
Guggulsterone enhances head and neck cancer therapies via inhibition of signal transducer and activator of transcription-3.
TLDR
The results suggest that guggulsterone-mediated inhibition of STAT3 and HIF-1alpha provide a biologic rationale for further clinical investigation of this compound in the treatment of HNSCC.
Guggulsterone-induced apoptosis in cholangiocarcinoma cells through ROS/JNK signaling pathway.
TLDR
The results showed that guggulsterone could induce apoptosis of human CCA cells through ROS/JNK signaling pathway, indicating that guGGulster one could be important for the clinical therapy of CCA.
Potential therapeutic targets of Guggulsterone in cancer
TLDR
Guggulsterone, a phytosterol isolated gum resin of the tree Commiphora mukul has been widely used in Indian traditional medicine as a remedy for various diseses and has been shown to possess cancer chemopreventive and therapeutic potential as established by in vitro and in vivo studies.

Guggulsterone-induced apoptosis in human prostate cancer cells is caused by reactive oxygen intermediate dependent activation of c-Jun NH2-terminal kinase.
TLDR
The present study reveals that the guggulsterone-induced cell death in human prostate cancer cells is regulated by ROI-dependent activation of JNK and guggursterone inhibits promoter activity of androgen receptor.
Curcumin Inhibits Hypoxia-Inducible Factor-1 by Degrading Aryl Hydrocarbon Receptor Nuclear Translocator: A Mechanism of Tumor Growth Inhibition
TLDR
It is found that curcumin inhibits Hif-1 activity and that this in turn down-regulates genes targeted by HIF-1, and the results suggest that the anticancer activity ofCurcumin is attributable to H IF-1 inactivation by ARNT degradation.
Targeting Stat3 blocks both HIF-1 and VEGF expression induced by multiple oncogenic growth signaling pathways
TLDR
Results presented here demonstrate that Stat3 is an effective target for inhibiting tumor VEGF expression and angiogenesis and regulates expression of Akt, which is required for growth signal-induced HIF-1 upregulation.
Regulation of colon carcinoma cell invasion by hypoxia-inducible factor 1.
TLDR
It is demonstrated that hypoxia or Hif-1 alpha overexpression stimulates Matrigel invasion by HCT116 human colon carcinoma cells, whereas this process is inhibited by a small interfering RNA directed against HIF-1alpha.
Caspase-dependent apoptosis induction by guggulsterone, a constituent of Ayurvedic medicinal plant Commiphora mukul, in PC-3 human prostate cancer cells is mediated by Bax and Bak
TLDR
It is indicated that caspase-dependent apoptosis by guggulsterone is mediated in part by Bax and Bak.

TLDR
VEGF can differentially induce STAT3 activation in micro‐ versus macro‐vascular endothelial cells and that this effect is linked to VEGFR2/STAT3 complex formation, which correlates with VEGF autocrine ability to stimulate its own gene expression.
Plant sterol guggulsterone inhibits nuclear factor‐&kgr;B signaling in intestinal epithelial cells by blocking I&kgr;B kinase and ameliorates acute murine colitis
TLDR
Guggursterone blocks NF‐&kgr;B signaling pathway by targeting IKK complex in IEC and attenuates DSS‐induced acute murine colitis, which suggests that guggulsterone could be an attractive therapeutic option in the treatment of IBD.
Constitutive Stat3 activity up-regulates VEGF expression and tumor angiogenesis
TLDR
It is shown that VEGF expression correlates with Stat3 activity in diverse human cancer cell lines and indicates that Stat3 represents a common molecular target for blocking angiogenesis induced by multiple signaling pathways in human cancers.
Nuclear Translocation of Phosphorylated STAT3 Is Essential for Vascular Endothelial Growth Factor-induced Human Dermal Microvascular Endothelial Cell Migration and Tube Formation*
TLDR
Analysis of the role of STAT3 in the migration and tube formation of the human dermal microvascular endothelial cells (HDMEC) and a dominant-negative STAT3 using adenovirus vector system demonstrate that STAT3 and its phosphorylation are involved in the downstream pathway of V EGF/VEGFR interaction and regulate VEGF-induced HDMEC migration andtube formation.
