}We previously reported association of eNOS with actin increases eNOS activity. In the present study, regulation of activity of eNOS by actin cytoskeleton during endothelial growth was studied. We found eNOS activity in PAEC increased when cells grew from preconfluence to confluence. eNOS activity was much greater in PAEC in higher density than those in lower density, suggesting increase in eNOS activity during cell growth is caused by increase in cell density. Although eNOS protein contents…

- Biology, ChemistryThe Journal of Biological Chemistry

The data indicate that increased association of eNOS with β-actin in PAEC contributes to hyperoxia-induced increase in the production of peroxynitrite which may cause nitrosative stress in pulmonary vasculature.

It is found that β-actin-induced increase in NO production was accompanied by decrease in superoxide formation, and this novel finding indicates that β -actin binding to eNOS through residues 326–333 in the eN OS protein results in shifting the enzymatic activity fromsuperoxide formation toward NO production.

Results suggested that stress fiber assembly, which controls EC reorientation and NO production, are dependent on rate and time of shearing, and there appear to be a relation between the cytoskeleton reorganization stage andNO production.

In endothelial cells, Nox4-derived H2O2 activates ERK1/2 to promote proliferation, whereas Nox2-containing NADPH oxidase maintains the cytoskeleton and prevents apoptosis to support cell survival, providing a new understanding of the molecular mechanisms that underpin endothelial cell survival.

ENOS is associated with actin in PAEC and that actin and its polymerization state play an important role in the regulation of eNOS activity is suggested.
Actin Cytoskeleton Organization and Posttranscriptional Regulation of Endothelial Nitric Oxide Synthase During Cell Growth

A role for actin cytoskeleton organization is identified in the posttranscriptional regulation of eNOS during cell growth and an inverse relationship between the concentration of G-actin and eNos expression was observed in endothelial cells subjected to pharmacological alteration of their cytos skeleton.

Analysis of the influence of cell-cell contact in the microvascular endothelial bEnd.3 cell line showed that eNOS activity was up-regulated in confluent monolayers compared with subconfluent cells, while there was no difference in eN OS expression, suggesting an association of eNos with the actin cytoskeleton.

The novel finding of this study is that the specific eNOS activity is upregulated in proliferating BAtEC and downregulated in quiescent B atrial endothelial cells, and the amount of "bioavailable" *NO is determined by eN OS activity and *NO inactivation (probably by superoxide), both high in proliferates BAt EC.
