Although glucosamine has been suggested to be effective in the treatment of osteoarthritis, its effect on disc degeneration remains unclear. We sought to explore whether glucosamine can activate autophagy in rat nucleus pulposus (NP) cells and protect cells treated with IL‐1β or hydrogen peroxide (H2O2). Autophagy in cells was examined by detecting for LC3, Beclin‐1, m‐TOR, and p70S6K, as well as by analyzing autophagosomes. To inhibit autophagy, 3‐methyladenine (3‐MA) was used. In the cells… Expand

Overexpressed TNFAIP3 can promote autophagy and reduce inflammation in LPS-induced human NPCs, providing a potential and an attractive therapeutic strategy for degenerative disease. Expand
Inhibiting IL‐1 signaling pathways to inhibit catabolic processes in disc degeneration
TLDR
Investigation of intracellular signaling pathways activated by IL‐1β, and GDF‐5 in the degenerate IVD to identify potential new therapeutic targets demonstrated p38 MAPK, c‐jun, and NFκB were activated during human IVD degeneration and inhibition of these pathways reduced or abrogated the catabolic effects of IL‐ 1β. MiR-210 facilitates ECM degradation by suppressing autophagy via silencing of ATG7 in human degenerated NP cells.
TLDR
It is suggested that miR-210 inhibits autophagy via silencing of ATG7, leading to increased Col II and aggrecan degradation in human degenerated NP cells. Expand

APO866 protects NP cells and induces autophagy by inhibiting IL-1β-induced NP cell degeneration and apoptosis, which may have therapeutic potential in IDD. Expand
Methylation of microRNA-129-5P modulates nucleus pulposus cell autophagy by targeting Beclin-1 in intervertebral disc degeneration
TLDR
It was found that miR-129-5P expression was downregulated while that of Beclin-1 and LC3-II was upregulated in degenerative disc NP cells, while autophagy was reduced in human NP cells transfected with miR -129-4P mimic or inhibitor, whereas the opposite result was observed upon treatment with miG3-3P inhibitor.
