Glucosamine and chondroitin sulfate, alone or in combination, are used worldwide by individuals suffering from osteoarthritis pain. They are by prescription in some countries but are available as over-the-counter dietary supplements in other countries, such as the United States. The inconclusive results of the National Institutes of Health-sponsored Glucosamine/chondroitin Arthritis Intervention Trial (GAIT) did little to clarify the efficacy of these agents. However, some newer studies have…

Glucosamine effects on lipids and blood pressure
TLDR
Past research studies on glucosamine, its efficacy in osteoarthritis, and the safety profile of the compound on lipids and blood pressure were inconclusive, even though they tended to suggest that glucosamines sulfate does not increase cholesterol levels; neither does it increase blood pressure in humans.
Oxaceprol Monotherapy versus Oxaceprol and Glucosamine Combination Therapy for Knee Osteoarthritis
TLDR
The efficacy of Oxaceprol and Glucosamine combination therapy is better than OxacEProl monotherapy, and further studies are required to examine mechanism of this effect at cellular level.
Short- and Long-Term Effectiveness of Supplementation with Non-Animal Chondroitin Sulphate on Inflammation, Oxidative Stress and Functional Status in Obese Subjects with Moderate Knee Osteoarthritis before and after Physical Stress: A Randomized, Double-Blind, Placebo-Controlled Trial
TLDR
In these obese adults with OA, nonanimal CS improved the inflammation, knee function, metabolic profile and body composition, and the result was significant in the CS group for BMI, WOMAC, CRP, total cholesterol and Homeostasis Model Assessment (HOMA).
Evaluation of a nutritional supplement for the alleviation of pain associated with feline degenerative joint disease: a prospective, randomized, stratified, double-blind, placebo-controlled clinical trial.
TLDR
This study showed a strong placebo effect and the glucosamine/chondroitin sulfate supplement did not show pain-relieving effects when compared with placebo, and no differences in activity were seen between the groups.
Marine Collagen Hydrolysates Promote Collagen Synthesis, Viability and Proliferation While Downregulating the Synthesis of Pro-Catabolic Markers in Human Articular Chondrocytes
TLDR
The results demonstrate that the use of Promerim® hydrolysates hold promise as complementary dietary supplements in combination with the current classical treatments or as a preventive therapy to delay the occurrence of OA in humans.
Joint Health
TLDR
Management strategies for arthritis range from lifestyle changes to invasive surgeries and also include topical treatments, physical therapy, non-steroidal antiinflammatory drugs, and injections.

The effect of glucosamine and/or chondroitin sulfate on the progression of knee osteoarthritis: a report from the glucosamine/chondroitin arthritis intervention trial.
TLDR
At 2 years, no treatment achieved a predefined threshold of clinically important difference in JSW loss as compared with placebo, however, knees with K/L grade 2 radiographic OA appeared to have the greatest potential for modification by these treatments.
In Vivo Chondroprotection and Metabolic Synergy of Glucosamine and Chondroitin Sulfate
TLDR
The data suggest that the disease modifying effect (the ability to retard progression of cartilage degeneration) of a mixture of glucosamine, chondroitin sulfate, and manganese ascorbate is more efficacious than either agent alone.
A pharmacoproteomic study confirms the synergistic effect of chondroitin sulfate and glucosamine
TLDR
The synergistic chondroprotective effect of CS and GlcN, first reported by this group at the intracellular level, is now demonstrated also at the extracellular level.
Randomized Trial of Glucosamine and Chondroitin Supplementation on Inflammation and Oxidative Stress Biomarkers and Plasma Proteomics Profiles in Healthy Humans
TLDR
A randomized, double-blind, placebo-controlled, cross-over study to assess the effects of glucosamine hydrochloride plus chondroitin sulfate for 28 days compared to placebo in healthy, overweight adults, adding evidence for potential mechanisms supporting epidemiologic findings that glucosamines and chond Detroitin are associated with reduced risk of lung and colorectal cancer.
Combined glucosamine and chondroitin sulfate, once or three times daily, provides clinically relevant analgesia in knee osteoarthritis
TLDR
This is a large study showing that GS/CS and GH/CS provide clinically meaningful and sustained analgesia in knee OA regardless of dose fractionation and capsule or sachet formulations.
Clinical efficacy and safety of glucosamine, chondroitin sulphate, their combination, celecoxib or placebo taken to treat osteoarthritis of the knee: 2-year results from GAIT
TLDR
Over 2 years, no treatment achieved a clinically important difference in WOMAC pain or function as compared with placebo, however, glucosamine and celecoxib showed beneficial but not significant trends.
Glucosamine and chondroitin for knee osteoarthritis: a double-blind randomised placebo-controlled clinical trial evaluating single and combination regimens
TLDR
Allocation to the glucosamine–chondroitin combination resulted in a statistically significant reduction in JSN at 2 years, and none of the treatment allocation groups demonstrated significant symptomatic benefit above placebo.
Combined chondroitin sulfate and glucosamine for painful knee osteoarthritis: a multicentre, randomised, double-blind, non-inferiority trial versus celecoxib
TLDR
CS+GH has comparable efficacy to celecoxib in reducing pain, stiffness, functional limitation and joint swelling/effusion after 6 months in patients with painful knee osteoarthritis, with a good safety profile.
Is All Glucosamine Alike? Clarifying the Controversies for Product Selection and Clinical Research
TLDR
This review describes the procedure that was developed by the authors to identify a high-quality glucosamine product for GAIT and to clarify confusing product information and nomenclature.
