β2-Microglobulin (β2m) is related to major histocompatibility complex class I alpha chains, and forms cell-surface glycoproteins that mediate a variety of functions in immune defense. In general, β2m has no isoforms and is not polymorphic in higher vertebrates, but polymorphisms between different alleles have been found in some fish species. In this study, full-length β2m cDNA and genomic sequences were cloned from the miiuy croaker (Miichthys miiuy). The miiuy croaker β2m gene shares many of…

The β2-Microglobulin Locus of Rainbow Trout (Oncorhynchus mykiss) Contains Three Polymorphic Genes1

Characterization of genomic clones from the same fish now shows that the rainbow trout β2m locus consists of two expressed genes and one partial gene that are closely linked, and a dramatic variation in the lengths of introns is caused by variable repetitive elements and accounts for the complex pattern seen in Southern blots.

Variation in the beta2m cDNA sequences is mainly at sites that do not perturb the structure of the maturebeta2m protein, showing that the observed diversity of the trout beta2M genes is not primarily a result of pathogen selection.

Cloning of the beta 2-microglobulin gene in the zebrafish.

The cDNA and genomic clones of the Brre-B2m gene are almost identical but the sequences of the 3'UT regions differ at 1.7% of the sites, suggesting that the genes borne by these clones might have diverged at least 0.7 million years ago.

Characterization of β2-microglobulin transcripts from two teleost species

The predicted protein secondary structure of both the carp and tilapia clones was almost identical to the corresponding regions of previously known vertebrate β2-microglobulin protein sequences, and Phylogenetic analysis indicated that the sequences were related to theβ2- microglobulins of higher vertebrates but grouped together in an ancestral position.

The human beta 2-microglobulin gene. Primary structure and definition of the transcriptional unit.

The nucleotide sequence of the human beta 2m gene and of a nearly full length cDNA clone are reported and it is revealed that exon/intron boundaries are absolutely conserved.
