Abstract.Chicken avidin and bacterial streptavidin, (strept)avidin, are proteins widely utilized in a number of applications in life science, ranging from purification and labeling techniques to diagnostics, and from targeted drug delivery to nanotechnology. (Strept)avidin-biotin technology relies on the extremely tight and specific affinity between (strept)avidin and biotin (dissociation constant, Kd≈10−14–10−16 M). (Strept)avidins are also exceptionally stable proteins. To study their ligand…

Bifunctional Avidin with Covalently Modifiable Ligand Binding Site

The bifunctional avidins described here provide improved and novel tools for applications such as the biofunctionalization of surfaces and could be covalently coupled to other maleimide derivatives, for instance fluorescent labels, allowing intratetrameric FRET.

Artificial Avidin-Based Receptors for a Panel of Small Molecules.

It is demonstrated that the avidin scaffold is suitable for use as a novel receptor for several biologically active small molecules and described the crystal structure of an antidin, sbAvd-2(I117Y), a steroid-binding avidin, which proves that the hypocrite scaffold can tolerate significant modifications without losing its characteristic tetrameric beta-barrel structure.

Trapavidin has the potential to replace streptavidin in many of its diverse applications, as well as providing an insight into the nature of ultra-stable noncovalent interactions.

Dual Chain Avidins – Useful Tools for Fusion Protein Applications

The ligand binding properties and structure of dual chain hybrid avidins (dchAvd) were investigated as well as to discuss their applicability in biotechnological applications to optimize the production and purification of these fusion proteins.

Chimeric avidin shows stability against harsh chemical conditions—biochemical analysis and 3D structure

It is found chimeric avidin to be more stable against various harsh organic solvents at elevated temperatures compared to avidin and streptavidin and to be a potential tool for new applications in biotechnology.

Chicken Avidin Exhibits Pseudo-catalytic Properties

The data demonstrate that several molecular features of avidin are responsible for the enhanced hydrolysis of biotinylp-nitrophenyl ester, including the nature of a decisive flexible loop, the presence of an obtrusive arginine, and a newly formed critical interaction between lysine 111 and the nitro group of the substrate.

Chicken avidin-related proteins show altered biotin-binding and physico-chemical properties as compared with avidin.

Differences were found in some physico-chemical properties of the AVRs as compared with avidin, including lowered pI, increased glycosylation and, most notably, reversible biotin binding for two AVs (AVR1 and AVR2), making the Avrs applicable for improved avidin-biotin technological applications.
