}The genetic content of wild-type human cytomegalovirus was investigated by sequencing the 235 645 bp genome of a low passage strain (Merlin). Substantial regions of the genome (genes RL1-UL11, UL105-UL112 and UL120-UL150) were also sequenced in several other strains, including two that had not been passaged in cell culture. Comparative analyses, which employed the published genome sequence of a high passage strain (AD169), indicated that Merlin accurately reflects the wild-type complement of…

Genetic Variability of Human Cytomegalovirus UL132 Gene in Strains from Infected Infants
- BiologyMicrobiology and immunology
- 2006
The results showed that a large number of nucleotide non‐synonymous substitutions occurred in the UL132 ORF, particularly in the 5′ half, in comparison to the UL 132 of reference strain, Toledo.
Sequential mutations associated with adaptation of human cytomegalovirus to growth in cell culture
- BiologyThe Journal of general virology
- 2010
Results suggest strongly that RL13 and UL128L exert at least partially independent suppressive effects on growth in fibroblasts, particularly those involved in cell tropism, immune evasion or growth temperance.
High-throughput sequence analysis of variants of human cytomegalovirus strains Towne and AD169
- BiologyThe Journal of general virology
- 2009
Using high-throughput technology, this work has sequenced HCMV strain Towne (ATCC VR-977) and confirmed the presence of two variants, one exhibiting the replacement in UL/b′ and the other intact in this region.
Genome sequences of human cytomegalovirus strain TB40/E variants propagated in fibroblasts and epithelial cells
- BiologyVirology journal
- 2021
The use of PacBio sequencing is reported to elucidate the genetic changes that occurred, both at the consensus level and within subpopulations, upon passaging a TB40/E stock on ARPE-19 epithelial cells and suggests that IE2, and perhaps its interactions with UL84, have important functions unique to HCMV replication in epithelial Cells.
Human cytomegalovirus UL145 gene is highly conserved among clinical strains
- BiologyJournal of Biosciences
- 2007
The structure of the UL145 gene and its putative protein are relatively conserved among clinical strains, irrespective of whether the strains come from patients with different manifestations, from different areas of the world, or were passaged or not in human embryonic lung fibroblast (HELF) cells.
Sequence variability of the α‐chemokine UL146 from clinical strains of human cytomegalovirus
- Biology
- 2004
Human cytomegalovirus (HCMV) is a ubiquitous pathogen that infects a variety of cell types in vivo. A region (referred to as UL/b') present in the Toledo strain of HCMV and low passage clinical…
Sequence Variability of Human Cytomegalovirus UL146 and UL147 Genes in Low-Passage Clinical Isolates

Sequence variability among HCMV clinical strains may affect the ability of UL146 and UL147 to attract human neutrophils and influence viral dissemination.

The coding content of clinical isolates is re-evaluated by identifying the set of open reading frames (ORFs) that are conserved in all five sequenced clinical isolate and which of these ORFs are present in the chimpanzee cytomegalovirus (CCMV) genome.
Genetic Stability of Bacterial Artificial Chromosome-Derived Human Cytomegalovirus during Culture In Vitro
- BiologyJournal of Virology
- 2016
Conditions under which BAC-derived viruses containing an intact, wild-type genome can be propagated in vitro with minimal risk of mutants being selected are identified, enabling studies of viruses expressing the gene complement of a clinical strain.
Reconstruction of the complete human cytomegalovirus genome in a BAC reveals RL13 to be a potent inhibitor of replication.
- BiologyThe Journal of clinical investigation
- 2010
To overcome rapid emergence of mutations in genetically intact HCMV, a system in which RL13 and UL131A were conditionally repressed during virus propagation is developed, which permits studies to be undertaken with a clonal, characterized H CMV strain containing the complete wild-type gene complement.

A dramatic level of genome sequence complexity is revealed that may explain the differences that these strains exhibit in virulence and tissue tropism and suggests that wild-type virus carries more than 220 genes, some of which are lost by large-scale deletion and rearrangement of the UL/b' region during laboratory passage.

The predicted UL144 product homology with tumor necrosis factor receptor family members, along with the unexpectedly high level of sequence variability of the UL144 ORF, suggests that the predicted product may play a role in HCMV infectivity and subsequent host disease.

An infectious bacterial artificial chromosome is used to introduce 413 defined insertion and substitution mutations into the human cytomegalovirus AD169 genome by random and site-directed transposon mutagenesis, producing mutations with a high probability of encoding proteins for which mutants have not been previously documented and in many previously characterized ORFs.
