Induced pluripotent stem (iPS) cells have been generated from mouse and human fibroblasts by the retroviral transduction of four transcription factors. However, the cell origins and molecular mechanisms of iPS cell induction remain elusive. This report describes the generation of iPS cells from adult mouse hepatocytes and gastric epithelial cells. These iPS cell clones appear to be equivalent to embryonic stem cells in gene expression and are competent to generate germline chimeras. Genetic… Expand
Induced Pluripotent Stem Cells Generated Without Viral Integration
TLDR
This work generated mouse induced pluripotent stem cells from fibroblasts and liver cells by using nonintegrating adenoviruses transiently expressing Oct4, Sox2, Klf4, and c-Myc, providing strong evidence that insertional mutagenesis is not required for in vitro reprogramming. Expand
Induced pluripotent stem cells generated without viral integration
TLDR
This work generates mouse induced pluripotent stem cells from fibroblasts and liver cells by using non-integrating adenoviruses transiently expressing Oct4, Sox2, Klf4, and c-Myc, providing strong evidence that insertional mutagenesis is not required for in vitro reprogramming. Expand
Recent advance in induced pluripotent stem cells
TLDR
The iPS cells will contribute to the fields ofelucidation of pathogenesis, drug discovery, toxicology study, and cell transplantation therapy in the future. Expand
Generation of Induced Pluripotent Stem (iPS) Cells by Nuclear Reprogramming
TLDR
Different aspects of nuclear reprogramming are discussed in this review article, different aspects of pluripotent stem cells can be derived from inner cell mass of blastocyte but its application is limited due to ethical concerns. Expand
Generation of mouse-induced pluripotent stem cells with plasmid vectors
TLDR
An alternative method to generate iPS cells from mouse embryonic fibroblasts (MEFs) by continual transfection of plasmid vectors is described, which reduces the safety concern for iPS cell generation and application, and provides a source of cells for the investigation of the mechanisms underlying reprogramming and pluripotency. Expand
Generation of induced pluripotent stem cells by efficient reprogramming of adult bone marrow cells.
TLDR
It is shown that mouse adult bone marrow mononuclear cells (BM MNCs) are competent as donor cells and can be reprogrammed into pluripotent ES cell-like cells, implying that BM M NCs have potential advantages to generate iPS cells for the clinical application. Expand
iPS cells produce viable mice through tetraploid complementation
TLDR
The generation of several iPS cell lines that are capable of generating viable, fertile live-born progeny by tetraploid complementation and confirm that iPS cells can attain true pluripotency that is similar to that of ES cells. Expand
Late Passage Human Fibroblasts Induced to Pluripotency Are Capable of Directed Neuronal Differentiation
TLDR
Successful reprogramming of human fibroblasts after more than 20 passages in vitro, to a pluripotent state with four transcription factors: Oct4, Sox2, Klf4, and c-Myc is reported. Expand
Reprogramming of Pancreatic β Cells into Induced Pluripotent Stem Cells
TLDR
The results provide genetic proof that terminally differentiated cells can be reprogrammed into pluripotent cells, suggesting that in vitro reprogramming is not restricted to certain cell types or differentiation stages. Expand
Variation in Hematopoietic Potential of Induced Pluripotent Stem Cell Lines
TLDR
It is concluded that hematopoietic potential differs among the six lines and that MEF-derived 20D17 and 178B5 iPS cells generate HCs more efficiently than adult–derived iPS cell lines. Expand

Generation of induced pluripotent stem cells without Myc from mouse and human fibroblasts

Induction of Pluripotent Stem Cells from Mouse Embryonic and Adult Fibroblast Cultures by Defined Factors
TLDR
Induction of pluripotent stem cells from mouse embryonic or adult fibroblasts by introducing four factors, Oct3/4, Sox2, c-Myc, and Klf4, under ES cell culture conditions is demonstrated and iPS cells, designated iPS, exhibit the morphology and growth properties of ES cells and express ES cell marker genes. Induced Pluripotent Stem Cell Lines Derived from Human Somatic Cells
Somatic cell nuclear transfer allows trans-acting factors present in the mammalian oocyte to reprogram somatic cell nuclei to an undifferentiated state. We show that four factors (OCT4, SOX2, NANOG,Expand
