The chemical tagging of a cytochrome P-450-dependent lauric acid omega-hydroxylase from clofibrate-treated Vicia sativa seedlings with [1-14C]11-dodecynoic acid allowed the isolation of a full-length cDNA designated CYP94A1. We describe here the functional expression of this novel P-450 in two Saccharomyces cerevisiae strains overproducing their own NADPH-cytochrome P-450 reductase or a reductase from Arabidopsis thaliana. The results show a much higher efficiency of the yeast strain…

Figures and Tables from this paper

Characterization of a methyl jasmonate and wounding‐responsive cytochrome P450 of Arabidopsis thaliana catalyzing dicarboxylic fatty acid formation in vitro
TLDR
A fatty‐acid‐metabolizing enzyme from Arabidopsis thaliana, CYP94C1, belonging to the cytochrome P450 family was cloned and characterized, revealing for the first time the presence of fatty acid in‐chain hydroxylase inArabidopsis.
Cloning and functional characterization of CYP94A2, a medium chain fatty acid hydroxylase from Vicia sativa.
TLDR
Heterologous expression of CYP94A2 in Saccharomyces cerevisiae yeast strain WAT11 shows that it catalyses the hydroxylation of myristic acid with a K(m(app)) of 4.0 microM and a turnover rate number of 80 min(-1).
CYP94A5, a new cytochrome P450 from Nicotiana tabacum is able to catalyze the oxidation of fatty acids to the omega-alcohol and to the corresponding diacid.
TLDR
A cytochrome P450 able to catalyze the complete set of reactions oxidizing a terminal methyl group to the corresponding carboxyl is described, which is enantioselective: after incubation of a synthetic racemic mixture of 9,10-epoxystearic acid, the chirality of the residual epoxide was 40/60 in favor of 9R,10S enantiomer.
CYP78A1 Preferentially Expressed in Developing Inflorescences of Zea mays Encoded a Cytochrome P450-Dependent Lauric Acid 12-Monooxygenase
TLDR
Based on results, CYP78A1 preferentially expressed in developing inflorescences of Zea mays appeared to have participated in the monooxygenation of fatty acids.
Molecular cloning of CYP76A3, a novel cytochrome P450 from Petunia hybrida catalyzing the ω-hydroxylation of myristic acid
TLDR
The reverse-transcriptase polymerase chain reaction was used to isolate a gene responsible for the oxidization of fatty acids from Petunia hybrida and a recombinant yeast microsome containing the CYP76A3 hemoprotein was found to specifically catalyze ω-hydroxylation of myristic acid.
Cloning, Functional Expression, and Characterization of CYP709C1, the First Sub-terminal Hydroxylase of Long Chain Fatty Acid in Plants
TLDR
CYP709C1 is the first identified plant cytochrome P450 that can catalyze sub-terminal hydroxylation of C18 fatty acids and postulated that it could be involved in plant defense by producing such compounds.
Molecular Cloning of CYP76B9, a Cytochrome P450 from Petunia hybrida, Catalyzing the ω-Hydroxylation of Capric Acid and Lauric Acid
TLDR
It is found that the ω-hydroxy metabolites of capric acid and lauric acid can affect the plant growth of Arabidopsis thaliana and be affected by preferential expression of the CYP76B9 gene in flowers and leaves.
CYP86B1 Is Required for Very Long Chain v-Hydroxyacid and a,v-Dicarboxylic Acid Synthesis in Root and Seed
TLDR
It is demonstrated that CYP86B1 is a very long chain fatty acid hydroxylase specifically involved in polyester monomer biosynthesis during the course of plant development.
CYP94A1, a plant cytochrome P450-catalyzing fatty acid ω-hydroxylase, is selectively induced by chemical stress in Vicia sativa seedlings
TLDR
The expression of CYP94A1 under these conditions and the concomitant morphological and cytological modifications would suggest the implication of this P450 in a process of plant defense against chemical injury.

TLDR
There appears to be as much divergence between plant families as there is between plant and animal, fungal or microbial families.
Induction of resistance in barley againstErysiphe graminisf.sp.hordeiby free cutin monomers
TLDR
Although the cutin monomers tested did not induce a number of defence-related barley genes, protection was eliminated by inhibiting host transcription with cordycepin, suggesting the involvement of inducible host reactions in the observed protection, and is compatible with a model that includes cut in monomers as signal molecules for the induction of disease resistance in barley.
