Polysulfated (oligo)flavonoids were synthesized and assayed for their in vitro and in vivo anticoagulant activities. The approach was based on molecular hybridization of two classes of anticoagulants, sulfated polysaccharides and sulfated flavonoids. The synthesis was optimized using microwave-assisted sulfation with triethylamine-sulfur trioxide. The obtained polysulfated flavonosides were highly effective in increasing clotting times and able to completely block the clotting process, in…

Polysulfated xanthones: multipathway development of a new generation of dual anticoagulant/antiplatelet agents.
TLDR
Dual anticoagulant/antiplatelet agents, such as sulfated xanthonosides 7 and 13, are expected to lead to a new therapeutic approach for the treatment of both venous and arterial thrombosis.
Coumarin derivatives from Ainsliaea fragrans and their anticoagulant activity
TLDR
Phytochemical investigation of the whole plant of Ainsliaea fragrans afforded five new coumarin derivatives, including a pair of natural 4-hydroxycoumarin enantiomers with a rare polycyclic pyrano[3-2c] carbon skeleton, together with 5 known biogenetically related compounds.
Dual anticoagulant/antiplatelet persulfated small molecules.
Sulfated Oligomers of Tyrosol: Toward a New Class of Bioinspired Nonsaccharidic Anticoagulants
TLDR
The preparation of sulfated derivatives of tyrosol, one of the most representative phenolic constituents of extra virgin olive oil, by different approaches shows how some structural facets of even closely related phenol polymers may be critical in dictating the anticoagulant activity, providing the key for the rationale design of active synthetic nonsaccharidic anticoaggulant agents alternative to heparin.

TLDR
This review highlights the synthetic approaches that were applied to obtain monosulfated or polysulfated phenolic small molecules and compiles the diverse biological activities already reported for this type of derivatives.
Searching for Small Molecules with Antioxidant and Anticoagulant Properties to Fight Thrombosis
TLDR
A new class of sulfated polyphenols derivatives which exhibited both in vitro and in vivo antithrombotic activity and antioxidant activity provided a model for the future design of dual anticoagulant-antioxidant agents.

Dual Anticoagulant/Antiplatelet Activity of Polyphenolic Grape Seeds Extract
TLDR
The results show, for the first time, the multi-potential effect of grape seed Extract on coagulation systems, and clearly suggest that grape seed extract could be considered a promising nutraceutical in the prevention of cardiovascular thrombotic events caused by different mechanisms.

Interaction of designed sulfated flavanoids with antithrombin: lessons on the design of organic activators.
TLDR
Competitive binding experiments indicate that activator CS does not compete with a saccharide ligand that binds antithrombin in the pentasaccharide binding site, while it competes with full-length low-affinity heparin.
The Unique Antithrombin III Binding Domain of Heparin: A Lead to New Synthetic Antithrombotics
TLDR
With the aid of molecular modeling and through “tailored” molecular modifications of the pentasaccharide, much knowledge has been gained concerning structure-activity relationships and more potent and simplified derivatives have been developed.

TLDR
The activation pattern of HAT-antibodies in the presence of linear curdlan sulfate fractions was dependent on the degree of sulfation and molecular weight of the glucan sulfates but not on the type of glycosidic linkage of a polysaccharide.

TLDR
DQD inhibited pig platelet aggregation induced by thrombin and its molecular mechanism was due to its inhibition ofCa2+ influx, intracellular Ca2+ mobilization, Ca2-/PL dependent PKC activity, and actin polymerization.
