[Objective] To establish a method for extraction and content determination of polysaccharide in Hedysayum polybotrys. [Method] Polysaccharide was extracted by water extraction and alcohol precipitation method. The content determination was performed with phenol- sulfuric acid as a chromomeric agent. [Result]The linear regression equation was Y = 0. 007X- 0. 013,R = 0. 999 6,indicating within the range of 0- 30 mg / L,the linear relationship between glucose concentration and absorbance was good. The content of polysaccharides in Hedysayum polybotrys,and HPS were respectively 4. 94%( RSD = 1. 06%),45. 308%( RSD = 1. 40%),and the average recovery was 99. 12%( RSD =0. 65%). [Conclusion] The method is simple,rapid,accurate,which could be used in extraction and content determination of Hedysayum polybotrys polysaccharide.
