Abstract The expression of the pyruvate kinase (PK) isoenzymes L and M2 was analysed in the livers of rats treated with the hepatocarcinogenic agent N-nitrosomorpholine (NNM) in the drinking water. In control animals L-PK expression was restricted to liver parenchymal cells, whereas M2-PK was detected in bile duct epithelial, blood vessel wall, endothelial and Kupffer cells. In rats treated with NNM proliferating oval cells were consistently L-PK negative and M2-PK positive, while the ductal… Expand
Tumor type M2 pyruvate kinase expression in gastric cancer, colorectal cancer and controls.
TLDR
Tumor M(2)-PK has a higher sensitivity than markers CEA and CA72-4, and is a valuable tumor marker for the detection of gastrointestinal cancer. Expand
Specific alterations of carbohydrate metabolism are associated with hepatocarcinogenesis in mitochondrially impaired mice.
TLDR
The hepatocarcinogenic process in AlbFxn(-/-) mice shows remarkable differences regarding carbohydrate metabolism alterations when compared with all other chemically and virally driven liver cancer models described up to now. Expand
The lack of evidence for correlation of pyruvate kinase M2 expression with tumor grade in non-small cell lung cancer.
BACKGROUND Over the last years, evidence has accumulated that an increased expression of pyruvate kinase M2 isozyme (PKM2) is related to neoplastic transformation as well that its plasmaExpand
Downregulation of TMEM70 in Rat Liver Cells After Hepatocarcinogen Treatment Related to the Warburg Effect in Hepatocarcinogenesis Producing GST-P-Expressing Proliferative Lesions
TLDR
A contribution of TMEM70 downregulation to the Warburg effect, which directs tumor promotion and progression in GST-P+-linked hepatocarcinogenesis in rats is suggested. Expand
Alterations in the glycolytic and glutaminolytic pathways after malignant transformation of rat liver oval cells
TLDR
Data from transformed oval cells are further proof that a special metabolic feature (increased glycolysis and glutaminolysis) is characteristic for tumor cells and that the mechanisms by which this metabolic state is induced can be totally different. Expand
Pyruvate Kinase M2 Expression, but Not Pyruvate Kinase Activity, Is Up-Regulated in a Grade-Specific Manner in Human Glioma
TLDR
It is shown that pyruvate kinase M expression, but not pyruVate Kinase activity, is regulated in a grade-specific manner in glioma, but that changes in both PK activity and PKM2 expression contribute to growth of GBM. Expand
INHIBITION OF EHRLICH ASCITES TUMOUR (EAT) CELLS PROLIFERATION THROUGH CHITOSAN-MEDIATED REGULATION OF ACTIVITY OF THE AKT PATHWAY
Isoenzyme M2 pyruvate kinase, which is a marker of cancer transformation, can take both tetramer (cytosol) and dimer (nucleus) forms. The former is responsible for ATP synthesis, and the latterExpand
Expression of pyruvate kinase M2 in human bladder cancer and its correlation with clinical parameters and prognosis
Background Pyruvate kinase M2 (PKM2) is a key regulator of the Warburg effect and has critical functions in glycolysis, contributing to the Warburg effect, tumor growth, angiogenesis, cell division,Expand
Faecal tumour M2 pyruvate kinase: a new, sensitive screening tool for colorectal cancer
TLDR
The determination of Tumour M2-PK in faeces provides a new promising screening tool for colorectal tumours and TNM and Dukes' classification of the tumours revealed a strong correlation between faecal Tumours M1-PK levels and staging. Expand
Response of sinusoidal mouse liver cells to choline-deficient ethionine-supplemented diet
TLDR
The results point to an important role of all types of sinusoidal cells in regeneration from CDE induced liver damage and call for utmost caution in using traditional marker for identifying specific cell types. Expand

Expression of L- and M2-pyruvate kinases in proliferating oval cells and cholangiocellular lesions developing in the livers of rats fed a methyl-deficient diet.
TLDR
This study clearly shows that the mere presence of a liver parenchymal cell marker in cells of the bile duct epithelial/oval cell compartment does not necessarily preclude that these cells are undergoing a differentiation into preneoplastic paren chymal cells, as has previously been suggested. Expand
Biochemical microanalysis of pyruvate kinase activity in preneoplastic and neoplastic liver lesions induced in rats by N-nitrosomorpholine.
TLDR
The results suggest that a reduction in PK activity is a relatively late event during the sequence of cellular changes leading from glycogenotic foci to hepatocellular carcinomas, which occurs only when low glycogen basophilic cell populations develop from the glycolysis and gluconeogenesis foci in later stages of hepatocarcinogenesis. Expand
Pyruvate kinase isoenzymes in altered foci and carcinoma of rat liver.
TLDR
The results do not support an isoenzyme shift from L to M2-PK in the course of malignant transformation of hepatocytes as suggested previously. Expand
Immunohistochemical demonstration of decreased L-pyruvate kinase in enzyme altered rat liver lesions produced by different carcinogens
TLDR
The results confirm the role of L-PK, an enzyme affecting the pentose phosphate pathway, as a negative marker of preneoplastic liver lesions in Wistar rats. Expand
Pyruvate kinase isoenzyme shift from L-type to M2-type is a late event in hepatocarcinogenesis induced in rats by a choline-deficient/DL-ethionine-supplemented diet.
TLDR
The results show that an isoenzyme shift from L- PK to M2-PK takes place at a late stage of the hepatocarcinogenic process, and that those MCF with a low L-PK expression and a reexpression of M2 -PK most probably represent the direct precursor lesions of hepatocellular neoplasms. Expand
Isozyme patterns of pyruvate kinase in various primary liver tumors induced during the process of hepatocarcinogenesis.
TLDR
Both nodular hyperplasia and highly differentiated hepatomas showed total activities similar to that of normal liver, while the proportion of M2-type activity was found to increase with statistical significance. Expand
Dual phenotypic expression of hepatocytes and bile ductular markers in developing and preneoplastic rat liver.
TLDR
It is demonstrated that oval and duct-like cells express both hepatocytic and bile ductular markers, and have the capacity to differentiate into hepatocytes, characteristics similar to hepatoblasts in the developing rat liver. Expand

Expression of alpha, mu and pi class glutathione S-transferases in oval and ductal cells in liver of rats placed on a choline-deficient, ethionine-supplemented diet.
TLDR
Immunocytochemical analysis revealed that the altered expression of GST in the whole liver is attributed to a differential expression of alpha, mu and pi class GSTs in the different cell types in the liver, including hepatocytes, oval cells around the portal region and among the sinusoids, and oval-like cells (ductal cells) in the 'duct-like' structures. Expand
Isozyme studies on adult, regenerating, precancerous and developing liver in relation to findings in hepatomas.
TLDR
Following controlled hyperplasia and differentiation after partial hepatectomy complete liver function is restored, whereas in the precancerous liver there is a loss of control of growth and differentiation in some cells resulting in hepatoma formation, which offers a rational explanation of the diverse phenotypes observed in the transplantable Morris liver hepatomas.
