Rabbit bladder epithelium, grown on collagen gels and exposed to the chemical carcinogen benzo[a]pyrene, produced nontumorigenic altered foci as well as tumorigenic epithelial cell lines during 120-180 d in culture. Immunofluorescence studies revealed extensive keratin filaments in both primary epithelial cells and benzo[a]pyrene-induced altered epithelial foci but showed no detectable vimentin filaments. The absence of vimentin expression in these cells was confirmed by two- dimensional gel…

Vimentin as a Marker of Early Differentiating, Highly Motile Corneal Epithelial Cells
- BiologyJournal of cellular physiology

Vim expression in corneal epithelium is found in a cell population composed of highly motile cells with a Vim+/Krt+/ΔNp63α+/Pax‐6low/α6 integrin+ phenotype, suggesting an indirect interaction between these proteins.
Epithelial cell migration requires the interaction between the vimentin and keratin intermediate filaments

It is demonstrated that vimentin co-localizes and co-immunoprecipitates with keratin-KRT14 and mutations in the -YRKLLEGEE- sequence of vimentsin significantly reduced migration of the keratinocytes.
Vimentin is necessary for colony growth of human diploid keratinocytes

It is suggested that Vim expression is essential for a highly motile phenotype, which is necessary for keratinocyte colony growth and possibly for development and wound healing, and long-term cultivation assays showed that Vim might be associated to pattern formation in cultured epithelia.

The results suggest that the extracellular matrix influences the cytoskeletal organization of the intermediate filaments and determines cell polarity, and confirm that collagen substrates permit epithelial cell cultures to progress toward a more differentiated state.

A method for initiating rapidly growing cultures of normal human transitional cells from ureter and embryonic bladder specimens has been developed and quantified and it was shown that cells growing out from the explants were continuous with the stratified urothelium maintained in the original explant.
Identification and Prognostic Significance of an Epithelial-Mesenchymal Transition Expression Profile in Human Bladder Tumors
- Medicine, BiologyClinical Cancer Research
- 2007
Altered markers of EMT defined within a panel of bladder carcinoma cell lines were recorded in vivo, frequently associated with tumors of high grade and stage and alterations associated with plakoglobin were identified as significant prognostic features in these tumors.
