Food effect (FE) and gastric pH-dependent drug-drug interactions (DDIs) are both absorption-related. Here, we evaluated if Biopharmaceutics Classification System (BCS) classes may be correlated with FE or pH-dependent DDIs. Trends in FE data were investigated for 170 drugs with clinical FE studies from the literature and new drugs approved from 2013 to 2019 by US Food and Drug Administration. Predicting Impact of Food and Feeding Time on Oral Absorption of Drugs with a Novel Rat Continuous Intestinal Absorption Model

Results indicate that the continuous intestinal rat model incorporates complex physiologic processes and feeding times relative to drug dose into a simple framework to provide accurate prediction of oral absorption.



Predicting Effect of Food on Extent of Drug Absorption Based on Physicochemical Properties

A logistic regression model based on dose, solubility, and permeability of compounds is developed to predict the food effect on AUC and it is found that for compounds with MAD ≥ clinical dose, thefood effect can be predicted from the dose number category and Log D category.

Relationship between the effects of food on the pharmacokinetics of oral antineoplastic drugs and their physicochemical properties

The results suggest that logP can be a useful parameter for predicting the effects of food on drug absorption, and drugs that will show increased absorption in the fed state and drugs for which absorption is not dependent on food can generally be predicted by logP.
