}The peritoneal exudate cells (PEC) represent the first line of defence against the virus in the mouse model of intraperitoneal (i.p.) infection with herpes simplex virus (HSV). We have therefore studied interferon production and activation of natural killer (NK) cells in vitro in PEC of HSV-injected mice. Injection of HSV caused a marked increase in NK cell activity, as reported by others. PEC from HSV-injected mice also produced high titres of interferon. This observation may be important…

Cultures of peritoneal exudate cells (PEC) of non-immune C57BL/6 mice reacted to infectious herpes simplex virus type 1 (HSV), but not to non-infectious virions, suggesting that in PEC the cells which produced interferon were of the macrophage lineage.

The data suggest that, in contrast to the situation observed with certain herpesviruses, interferon and NK cells may not be of overwhelming importance in the defense of mice against MHV3.

Inbred mouse strains differ in susceptibility to infection with herpes simplex virus type 1 or type 2 (HSV-1, HSV-2), and early (2 to 4 hours) interferon production was correlated to resistance.

Clinical observations suggest that during recurrent HSV infection IFN γ is secreted locally by activated T lymphocytes and may synergize with IFN α/β in the antiviral activity against HSV.

Experiments with silica-treated mice, and with homozygous nude mice, suggested that the observed restriction in virus transfer is independent of T-cell and macrophage functions, suggesting that age-dependent defence mechanisms are of relevance in the restriction of peripheral infection by HSV-1 ANG in adult mice.

Newborn C57BL/6 mice, in contrast to adult mice, did not develop natural killer cell activity in the peritoneal cavity 24 h after i.p. injection of HSV-1, and showed only minimal titres of interferon in thePeritoneal fluid after 4 h.

An analysis of virus-specified macromolecular syntheses in infected macrophages indicated that the major viral-specific proteins were induced in macrophage cultures, and it is suggested that an error in DNA metabolism may be the primary cause of restriction.

The distinction between NK cells elicited in the spleen and peritoneal cavity of mice inoculated intravenously or intraperitoneally with live Kunjin virus and NK cells induced by either agent suggests that there may be more than one type of NK cell or that activation enhances the cytotoxicity and perhaps broadens the range of target specificity of endogenous NK cells.
