We measured the genotoxic activities in two bacterial tests, the Salmonella/histidine assay (a reverse mutation assay) and the SOS chromotest (an assay for SOS induction in E. coli), of three 2-nitroanthrafurans: 2-nitroanthra[1,2-b]furan (R-7688), the isomeric compound 2-nitroanthra[2,1-b]furan (R-7686) and its 8-methoxylated derivative (R-7707). Their genotoxic activities were compared to that of 7-methoxy-2-nitronaphtho[2,1-b]furan (R-7000) which has been studied in previous works (Arnaise… Expand

The structural basis of the genotoxicity of nitroarenofurans and related compounds.
Using CASE in the predictive mode, the program correctly predicted the probability of sfiA induction in E. coli of 95.8% of 24 "unknown" nitroarenofurans which were not part of the learning set (QSAR with r = 0.88-0.97). Expand
The SOS chromotest: a review.
The SOS chromotest is one of the most rapid and simple short-term test for genotoxins and is easily adaptable to various conditions, so that it could be used as an early--perhaps the earliest--test in a battery. Expand
SOS chromotest results in a broader context: Empirical relationships between genotoxic potency, mutagenic potency, and carcinogenic potency
This work investigated the empirical relationships between SOS genotoxic potency and mutagenic potency (as measured with the Salmonella/microsome assay), as well as between genot toxic potency and carcinogenic potency ( as measured using standard, chronic animal bioassays). Expand

Genotoxic activity of two furan analogues of benzo[a]pyrene and their 2-nitro derivatives.
The responses to the two pyrenofurans were increased in the presence of an "activating mixture" but were still lower than that to benzo[a]pyrene and pyreno[1,2-b]furan (R7490), and compounds with the oxygen heteroatom within the "bay region" gave lower responses than their isomeric furan analogues outside the "Bay region". Genotoxic activities of 2-nitronaphthofurans and related molecules.
The genotoxic activities of 63, 2-nitronaphthofurans and related molecules were examined using two bacterial short-term tests, the Salmonella mammalian microsome assay test or Mutatest, a mutagenesisExpand
Mutagenic activity of benzofurans and naphthofurans in the Salmonella/microsome assay: 2-nitro-7-methoxy-naphtho[2,1-b]furan (R7000), a new highly potent mutagenic agent.
One of the compounds tested, 2-nitro-7-methoxynaphtho[2,1-b]furan (R7000), may be the most potent mutagen examined so far in the Salmonella/microsome assay and depends notably on the nature and the position of the other substituents in the molecule. Expand
Relationship between the chemical structure and the mutagenic and carcinogenic potentials of five naphthofurans.
The positive results obtained in the cell transformation assays and in the short-term skin tests indicate that Compounds A, B, and C are probably carcinogenic, and further in vivo studies should be accomplished before using the 2-nitronaphthofuran derivatives in human and animal treatments. Expand
Genetic toxicology studies with 2-nitrobenzofurans and 2-nitronaphthofurans.
A correlation is observed between the mutagenic potency assayed in the Mutatest and the phage-inducing potency assays in the Inductest, and the relevance of complementary tests on uvr + Salmonella strains and the Spermatest are briefly discussed. Expand
SOS chromotest, a direct assay of induction of an SOS function in Escherichia coli K-12 to measure genotoxicity.
The results suggest that mutagenic potency measured in the mutatest reflects the level of induction of an SOS function and that most genotoxins are inducers of the SOS response in bacteria. Expand
Synthesis, spectral analysis, and mutagenicity of 1-, 3-, and 6-nitrobenzo[a]pyrene.
Comparison of the spectral data of these compounds with those obtained from direct nitration of benzo[a]pyrene confirmed that 1- and 3-nitrobenzo[ a]pyrenes are indeed the minor products of the latter reaction, and verifies that the minor nitrated products of benzos[a ]pyrene formed in model air atmospheres. Induction of sarcomas in rats by subcutaneous injection of 7-methoxy-2-nitro-naphtho[2,1-b]furan (R 7000).
The high carcinogenicity of R 7000 in vivo is analogous to that of methylcholanthrene, and the exact mechanism of action of 2-nitro-naphthofurans as carcinogens remains to be explained. Expand
