The ability of metals to activate estrogen receptor-alpha (ERalpha) was measured in the human breast cancer cell line, MCF-7. Similar to estradiol, treatment of cells with the divalent metals copper, cobalt, nickel, lead, mercury, tin, and chromium or with the metal anion vanadate stimulated cell proliferation; by d 6, there was a 2- to 5-fold increase in cell number. The metals also decreased the concentration of ERalpha protein and mRNA by 40-60% and induced expression of the estrogen…

Figures and Tables from this paper

Activation of estrogen receptor-alpha by the anion nitrite.
TLDR
Concentrations of nitrite induced a significant increase in PgR mRNA, suggesting that physiologically and environmentally relevant doses of the anion activate ERalpha, and diphenyleneiodonium was used to inhibit oxidation/reduction reactions in the cell.
Arsenite and cadmium promote the development of mammary tumors
TLDR
It was shown that environmentally relevant amounts of arsenite and cadmium had estrogen-like activity in vivo and promoted mammary tumorigenesis.
Cadmium induces mitogenic signaling in breast cancer cell by an ERα-dependent mechanism
Effects of Tobacco Smoke Condensate on Estrogen Receptor-α Gene Expression and Activity
TLDR
Investigating whether tobacco smoke condensate (TSC), which contains metals and metalloids, elicits estrogen-like effects at environmentally relevant doses, results in a 2.5-fold stimulation of cell growth in human breast cancer cells.
Induction of Hsp22 (HspB8) by estrogen and the metalloestrogen cadmium in estrogen receptor– positive breast cancer cells
TLDR
Hsp22 induction represents a new aspect of the estrogenic response with potential significance for the biology of estrogen receptor– positive breast cancer cells.
Transcriptional effects of metal ions on the bovine oxytocin and the thymidine kinase-ERE promoter through the estrogen receptor α in MDA-MB 231 breast cancer cell line
TLDR
Investigation of the Transcriptional effects of variety of metal ions on the bovine oxytocin and the thymidine kinase-ERE promoter by estrogen receptor α in MDA-MB 231 breast cancer cell line revealed that some metal ions show estrogenic activity by classical or non-classical mechanisms as well as someMetal ions exhibit estrogenicactivity by undetermined mechanisms in transfected MDA -MB 231 cell line.
Arsenic-induced cancer cell phenotype in human breast epithelia is estrogen receptor-independent but involves aromatase activation
TLDR
Chronic arsenic exposure drives human breast epithelia into a cancer cell phenotype with an apparent overabundance of putative CSCs, thereby activating oncogenic processes independent of ER.
Estrogenic activity of cadmium, copper and zinc in the yeast estrogen screen.
Differential Effects of Organic and Inorganic Mercury on PhenotypicallyVariant Breast Cancer Cell Lines
TLDR
Protective effects were not observed in the tumor cell lines with the MCF-7 (ER+) cells being the most resistant to the mercury effects with potential involvement of this receptor in the effects of mercuric compounds.

Effects of Arsenite on Estrogen Receptor-α Expression and Activity in MCF-7 Breast Cancer Cells.
TLDR
The effects of arsenite on the chimeric receptor were blocked by the antiestrogen, suggesting that arsenite activates ERalpha through an interaction with the hormone-binding domain of the receptor.
Effect of cadmium on estrogen receptor levels and estrogen-induced responses in human breast cancer cells.
TLDR
The data suggest that the effects of cadmium are mediated by the estrogen receptor independent of estradiol, and Cadmium induced the growth of MCF-7 cells 5.6-fold and pS2 and progesterone receptor mRNAs in transient transfection experiments.
Activation of estrogen receptor-alpha by the heavy metal cadmium.
TLDR
The ability of cadmium to bind to and activate wild-type and various mutants of ER-alpha was examined, suggesting that the heavy metal interacts with the hormone-binding domain of the receptor.
Activation of Estrogen Receptor-α by the Heavy Metal Cadmium
TLDR
The ability of cadmium to bind to and activate wild-type and various mutants of ER-alpha was examined, suggesting that the heavy metal interacts with the hormone-binding domain of the receptor.
Effects of selenite on estrogen receptor‐α expression and activity in MCF‐7 breast cancer cells
TLDR
The effects of selenite on the chimeric receptor were blocked by the antiestrogen, suggesting that selenites activates ER‐α through an interaction with the hormone binding domain of the receptor.
Regulation of the estrogen receptor in MCF-7 cells by estradiol.
TLDR
The data suggest that estrogen down-regulates ER mRNA by inhibition of ER gene transcription at early times and by a posttranscriptional effect on receptor mRNA at later times.
Effect of metal ions on the binding of 17beta-estradiol to human endometrial cytosol.
TLDR
Analysis of the binding data revealed that one possible site of action of the cations on the binding protein might be the sulfhydryl group(s) of the 17beta-estradiol binding site, which was found to be the most potent inhibitor.
Effects of copper on cytoplasmic and nuclear binding of 17 beta-estradiol in the rat uterus.
TLDR
Results show that Cu++ can inhibit [3H]E2 binding to tissue cytoplasmic receptors in vitro and thereby interfere with hormone delivery to target cell nuclei.
