OBJECTIVE To assess the effect of oral glucose loading on plasma endothelin-1 (ET-1) levels in humans. RESEARCH DESIGN AND METHODS A total of 75 g D-glucose was given orally to 14 nonobese nondiabetic essential hypertensive subjects (eight men and six women, mean age 43.1 ± 3.0 years) and eight normotensive subjects (four men and four women, mean age 45.2 ± 4.1 years). Blood samples for plasma ET-1 measurement were drawn every 30 min for 2 h and then at 180 and 240 min. RESULTS After glucose…

Endothelin‐1 infusion inhibits plasma insulin responsiveness in normal men
TLDR
It is demonstrated that an increase in circulating ET-1 to levels observed in insulin-resistant states acutely diminishes the insulin secretory response but does not significantly modify insulin sensitivity.
Insulin infusion induces endothelin-1-dependent hypertension in rats.
TLDR
The results suggest that, in the insulin infusion rat model, ET-1 plays a mediating role in the development of hypertension, but not of insulin resistance.
Effects of Hyperinsulinemia on Blood Pressure in High-Fat Diet Fed Rats
TLDR
Hyperinsulinemia following high-fat diet is associated with an elevation in blood pressure and sodium retention, increased plasma endothelin-1 and noradrenaline, as well as activation of renin-angiotensin system may all contribute to the blood pressure elevation.
Hyperinsulinemia fails to augment ET-1 action in the skeletal muscle vascular bed in vivo in humans.
TLDR
Endothelin antagonism augmented insulin-stimulated NO bioavailability and NOx flux, but not differently between groups and not proportional to hyperinsulinemia, and these findings do not support the hypothesis that insulin resistance-associated hyperinsulainemia preferentially drives endothelin-mediated vasoconstriction.
Endothelial Activation in Patients With Cardiac Syndrome X
TLDR
The findings show that no basal endothelial damage is present in patients with cardiac syndrome X, and increased responsiveness of endothelin-1 to glucose loading suggests that patients with heart syndrome X present an increased susceptibility to releasing endothelins-1 under stressful circumstances.
Endothelial Activation in Patients With Cardiac Syndrome
TLDR
The findings show that no basal endothelial damage is present in patients with cardiac syndrome X, and increased responsiveness of endothelin-1 to glucose loading suggests that patients with heart syndrome X present an increased susceptibility to releasing endothelins-1 under stressful circumstances.

Plasma Endothelin-1 Levels in Obese Hypertensive and Normotensive Men
TLDR
It is shown that plasma ET-1 concentrations are increased in human obesity, which could depend on fasting insulin concentrations, abnormal peptide clearance, or both, and Hypertension does not influence the behavior of plasmaET-1 levels in obese men.
Elevated Endothelin‐1 in Heart Failure and Loss of Normal Response to Postural Change
TLDR
Alterations in plasma levels of endothelin in congestive heart failure and in response to postural change were qualitatively and quantitatively similar to the alterations of known mediators of neurohumoral compensation.

Insulin stimulates endothelin binding and action on cultured vascular smooth muscle cells.
TLDR
It is postulate that hyperinsulinemia in vivo may potentiate ET release and receptor-mediated action, thereby contributing to vascular disease in the setting of diabetes.

Endothelin, a vasoconstrictor peptide secreted from endothelial cells, has been thought to play a role in various forms of vascular disease. Diabetes mellitus is well known for its association with
Assessment of insulin sensitivity in vivo.
TLDR
Evidence emerged suggesting that human diabetes mellitus has a multifactorial etiology, and larger doses of insulin were required to normalize the blood sugar in patients with the milder nonketotic form of the disease common in the older population, whereas smaller doses were adequate for younger, ketosis-prone diabetics.
