OBJECTIVE To assess the effect of oral glucose loading on plasma endothelin-1 (ET-1) levels in humans. RESEARCH DESIGN AND METHODS A total of 75 g D-glucose was given orally to 14 nonobese nondiabetic essential hypertensive subjects (eight men and six women, mean age 43.1 ± 3.0 years) and eight normotensive subjects (four men and four women, mean age 45.2 ± 4.1 years). Blood samples for plasma ET-1 measurement were drawn every 30 min for 2 h and then at 180 and 240 min. RESULTS After glucose…

It is demonstrated that an increase in circulating ET-1 to levels observed in insulin-resistant states acutely diminishes the insulin secretory response but does not significantly modify insulin sensitivity.

The results suggest that, in the insulin infusion rat model, ET-1 plays a mediating role in the development of hypertension, but not of insulin resistance.

Effects of Hyperinsulinemia on Blood Pressure in High-Fat Diet Fed Rats
- Biology, Medicine
- 2016
Hyperinsulinemia following high-fat diet is associated with an elevation in blood pressure and sodium retention, increased plasma endothelin-1 and noradrenaline, as well as activation of renin-angiotensin system may all contribute to the blood pressure elevation.
Hyperinsulinemia fails to augment ET-1 action in the skeletal muscle vascular bed in vivo in humans.

Endothelin antagonism augmented insulin-stimulated NO bioavailability and NOx flux, but not differently between groups and not proportional to hyperinsulinemia, and these findings do not support the hypothesis that insulin resistance-associated hyperinsulainemia preferentially drives endothelin-mediated vasoconstriction.

Endothelial Activation in Patients With Cardiac Syndrome X
- Medicine, BiologyCirculation

The findings show that no basal endothelial damage is present in patients with cardiac syndrome X, and increased responsiveness of endothelin-1 to glucose loading suggests that patients with heart syndrome X present an increased susceptibility to releasing endothelins-1 under stressful circumstances.

It is shown that plasma ET-1 concentrations are increased in human obesity, which could depend on fasting insulin concentrations, abnormal peptide clearance, or both, and Hypertension does not influence the behavior of plasmaET-1 levels in obese men.
