Heightened interest in relevant models for human disease increases the need for improved methods for germline transgenesis. We describe a significant improvement in the creation of transgenic laboratory mice and rats by chemical modification of Sleeping Beauty transposons. Germline transgenesis in mice and rats was significantly enhanced by in vitro cytosine-phosphodiester-guanine methylation of transposons prior to injection. Heritability of transgene alleles was also greater from founder mice…
Germline Transgenic Pigs by Sleeping Beauty Transposition in Porcine Zygotes and Targeted Integration in the Pig Genome
TLDR
Transposase-catalyzed transgenesis in a large mammalian species expands the arsenal of transgenic technologies for use in domestic animals and will facilitate the development of large animal models for human diseases.
Strategies for selection marker-free swine transgenesis using the Sleeping Beauty transposon system
TLDR
It is demonstrated that the Sleeping Beauty (SB) transposon system effectively delivers monomeric, multi-copy transgenes to the pig embryo genome by pronuclear injection without markers, as well as to donor cells for founder generation by cloning.
Transposon‐mediated transgenesis, transgenic rescue, and tissue‐specific gene expression in rodents and rabbits
TLDR
A side‐by‐side comparison of the SB100X‐ and piggyBac‐based protocols revealed that the two systems are complementary, offering new opportunities in genome manipulation.
Assessing Tn5 and Sleeping Beauty for transpositional transgenesis by cytoplasmic injection into bovine and ovine zygotes
TLDR
Results show that transpositional transgenesis by cytoplasmic injection of Tn5 and Sleeping Beauty transposon components can be applied for the production of transgenic lambs of pharmaceutical interest.
Remobilization of Sleeping Beauty transposons in the germline of Xenopus tropicalis
TLDR
It is demonstrated that SB transposons integrated into the X. tropicalis genome are effective substrates for excision and re-integration, and that the remobilized transposon are transmitted through the germline.
Novel strategies for gene trapping and insertional mutagenesis mediated by Sleeping Beauty transposon
TLDR
Two novel trapping vectors mediated by Sleeping Beauty transposon are generated by the combination of three functional cassettes that are required for finding endogenous genes, disrupting the expression of trapped genes, and inducing the excision of integrated traps from their original insertion sites and then inserting into another gene.
Assessment of Fecundity and Germ Line Transmission in Two Transgenic Pig Lines Produced by Sleeping Beauty Transposition
TLDR
The results show that Sleeping Beauty transposase-catalyzed transposition is a promising approach for stable genetic modification of the pig genome.
Generation of minipigs with targeted transgene insertion by recombinase-mediated cassette exchange (RMCE) and somatic cell nuclear transfer (SCNT)
TLDR
The results show that RMCE can be done in a pre-selected transcriptionally active acceptor locus for targeted transgenesis in piglets with an expression profile in various tissues similar to that of the GFP marker gene.

DNA methylation of Sleeping Beauty with transposition into the mouse genome
TLDR
The results indicate that transposition via Sleeping Beauty into the mouse genome may result in a significant level of de novo DNA methylation, which may depend on a number of different factors including the cargo DNA sequence, chromosomal context of the insertion site, and/or host genetic background.
Conditional gene expression in the mouse using a Sleeping Beauty gene-trap transposon

Molecular evolution of a novel hyperactive Sleeping Beauty transposase enables robust stable gene transfer in vertebrates
TLDR
The newly developed transposase yields unprecedented stable gene transfer efficiencies following nonviral gene delivery that compare favorably to stable transduction efficiencies with integrating viral vectors and is expected to facilitate widespread applications in functional genomics and gene therapy.

Characterization of Sleeping Beauty Transposition and Its Application to Genetic Screening in Mice
TLDR
Transposition sites not only were clustered within 3 Mb near the donor site but also were widely distributed outside this cluster, indicating that the SB transposon can be utilized for both region-specific and genome-wide mutagenesis.

Mammalian germ-line transgenesis by transposition
TLDR
Experiments in which transposon vectors were injected into one-cell mouse embryos with mRNA expressing the SB10 transposase enzyme showed promise as a germ-line transgenesis method in other vertebrate species and for insertional mutagenesis in the mouse.
