Although local anesthetics (LA) are considered primarily Na+-channel blockers in the past decade, an alternative action of LA as inhibitors of fusion among compartments of the endocytic/exocytic pathways was described. In epimastigote forms of Trypanosoma cruzi, we observed that 50 mM dibucaine reduced the rates of uptake of bovine serum albumin (BSA) and immunoglobulin to 60% of control values in addition to the delay of exocytosis of cysteine proteases. Fusion among endocytic compartments was… Expand
Endocytosis and Exocytosis in Leishmania amazonensis Are Modulated by Bromoenol Lactone
The results suggested that the activity inhibited by BEL, which is one of the irreversible inhibitors of iPLA2, is required for both endocytosis and exocytotic in promastigotes of L. amazonensis. Expand
Trypanosoma cruzi Peptidases: An Overview
The main T. cruzi peptidases are examined, focusing on their biological roles, especially concerning the parasite-mammalian host relations. Expand
Synthesis and In Vitro Antibacterial Evaluation of Schiff Bases Derived FROM 2-Chloro-3-Quinolinecarboxaldehyde
Background: Design, identification, and synthesis of new antimicrobial agents along with preventive proceedings are essential to confront antibiotic-resistant pathogenic bacteria. HeterocyclicExpand

Identification of a large pre-lysosomal compartment in the pathogenic protozoon Trypanosoma cruzi.
Immunocytochemical data from Lowicryl-embedded cells demonstrated that the reservosomes are acidic compartments with no acid phosphatase or typical lysosome-associated membrane proteins, but rich in cysteine proteinase, suggesting thatThe reservosome is a pre-lysosomal compartment. Expand
Developmental and morphological regulation of clathrin-mediated endocytosis in Trypanosoma brucei.
Major endocytic coat proteins in T. brucei are described for the first time, andModulation of clathrin expression is likely to be an important factor in the developmental regulation of endocytosis and recycling in the African trypanosome. Expand
Cysteine protease inhibitors alter Golgi complex ultrastructure and function in Trypanosoma cruzi.
It is suggested that cysteine protease inhibitors prevent the normal autocatalytic processing and trafficking of cruzain within the Golgi apparatus. Expand
Endocytosis of gold-labeled proteins and LDL by Trypanosoma cruzi.
Endocytosis was studied at the ultrastructural level in different developmental forms of Trypanosoma cruzi after incubation of the parasites in the presence of gold-labeled proteins (albumin-Au,Expand
Trypanosoma cruzi: ultrastructural, cytochemical and freeze-fracture studies of protein uptake.
The cell membrane as well as the membranes of the pinocytotic vesicles and the large multivesicular structure have carbohydrates, as detected by the periodic acid-thiosemicarbazide-silver proteinate technique. Expand
Receptor-mediated endocytosis in the bloodstream form of Trypanosoma brucei.
The uptake of various host plasma proteins by the bloodstream form of Trypanosoma brucei was studied both biochemically, using radiolabeled proteins, and with the electron microscope, using colloidalExpand
Characterization of a Rab11 homologue in Trypanosoma cruzi.
A Trypanosoma cruzi Rab 11 homologue is characterized that shares at, the amino acid level, 40% similarity with human rab11, Arabdopsis thaliana rab11 and yeast rab11 homologue genes and may represent the first molecular marker of this peculiar organelle of Vesicle trafficking between organelles. Expand
Characterization of a Rab11 homologue in Trypanosoma cruzi.
Interestingly, the TcRab11 product seems to be located at the reservosome complex, a site of active endocytosis and vesicle fusion present only in the epimastigote stage, and may represent the first molecular marker of this peculiar organelle. Expand
Low temperature blocks fluid-phase pinocytosis and receptor-mediated endocytosis in Trypanosoma cruzi epimastigotes
The results suggest that low temperatures affect the transport and pinching of endocytic vesicles as well as the rate of delivery of transferrin to reservosomes. Expand
Cysteine proteinase in Trypanosoma cruzi: immunocytochemical localization and involvement in parasite-host cell interaction.
A monospecific polyclonal antibody obtained against a cysteine proteinase isolated from epimastigotes of Trypanosoma cruzi was used for the immunocytochemical localization of the protein by electronExpand
