“Decoy” oligonucleotides can be used as gene‐specific nuclear factor (NF‐κB) inhibitors to regulate gene expression. We applied two different decoy oligonucleotides that contained the NF‐κB binding consensus sequences present in the immunoglobulin G (IgG)‐κB and Bcl‐x promoter into 7‐day‐old (P7) rat lateral ventricles before hypoxia/ischemia (HI) and compared their effects on gene expression in hippocampi to saline‐treated, scrambled decoy‐treated, or untreated hippocampi exposed to HI. A decoy oligonucleotide inhibiting nuclear factor‐κB binding to the IgGκB consensus site reduces cerebral injury and apoptosis in neonatal hypoxic‐ischemic encephalopathy
TLDR
It is likely that the decoy oligonucleotide affects cerebral injury and apoptosis not through suppression of downstream elements of the inflammatory response but through other mechanisms, one of which is the reduction of transcription and synthesis of cytokines, which are known to affect other responses to cellular injury.
Nuclear factor‐κB decoy amelioration of spinal cord injury‐induced inflammation and behavior outcomes
TLDR
NF‐κB p65/p50 targeted decoys improved early locomotor recovery after moderate but not severe SCI, yet ameliorated SCI‐induced hypersensitization after both moderate and severeSCI.
Differential regulation of BACE1 promoter activity by nuclear factor‐κB in neurons and glia upon exposure to β‐amyloid peptides
TLDR
A signal transduction pathway is characterized that may be responsible for the increases in Aβ associated with AD, and it is proposed that the transcription factor NF‐κB acts as a repressor in neurons but as an activator of BACE1 transcription in activated astrocytes present in the CNS under chronic stress, a featurepresent in the AD brain.
Activation of nuclear factor‐κB signaling pathway by interleukin‐1 after hypoxia/ischemia in neonatal rat hippocampus and cortex
Perinatal hypoxia/ischemia (HI) is a common cause of neurological deficits in children. Interleukin‐1 (IL‐1) activity has been implicated in HI‐induced brain damage. However, the mechanisms
Sequestration of serum response factor in the hippocampus impairs long‐term spatial memory
TLDR
It is found that intra‐hippocampal infusion of NFkappaB decoy oligonucleotides also impairs long‐term spatial memory, consistent with a role for this factor in long-term memory storage, and suggest that SRF‐mediated gene expression is required for long‐ term spatial memory.
The transcription factor Yin Yang 1 is an activator of BACE1 expression
TLDR
It is shown that Yin Yang 1 (YY1), a highly conserved and multifunctional transcription factor, binds to its putative recognition sequence within the BACE1 promoter and stimulates Bace1 promoter activity in rat pheochromocytoma 12 cells, rat primary neurones and astrocytes.
Does Inhibition of Nuclear Factor kappa B Explain the Protective Effect of Ticagrelor on Myocardial Ischemia Reperfusion Injury?
TLDR
The article “Ticagrelor Reduces Ischemia-Reperfusion Injury Through the NFkB–Dependent Pathway in Rats” by Liu et al was puzzled by the minimal number of cytokines tested and the reliance on reverse transcription polymerase chain reaction and Western blots for total p65 as indices of NFkB activity.
Enhancing Expression of Nrf2-Driven Genes Protects the Blood–Brain Barrier after Brain Injury
TLDR
Findings indicate a potential therapeutic usefulness for Nrf2-activating molecules to improve the function of the neurovascular unit after injury.
Short and long TNF‐alpha exposure recapitulates canonical astrogliosis events in human‐induced pluripotent stem cells‐derived astrocytes
TLDR
The results indicate that human iPSC‐derived astrocytes reproduce canonical events associated with astrogliosis in a time dependent fashion, which may contribute to a better understanding of mechanisms governing human astrogLiosis.
Perinatal brain injury and regulation of transcription
TLDR
Investigation of the transcriptional mechanisms of neuro-survival is likely to reveal other novel transcription factors whose activation by small molecules or drugs will complement current medication in activating the salutary gene program.

Differential NF‐κB regulation of bcl‐x gene expression in hippocampus and basal forebrain in response to hypoxia
TLDR
There is a tissue‐specific regulation of bcl‐x gene expression by NF‐κB in hypoxia‐induced cell death in the hippocampus, suggesting that the absence of these regulating features in the basal forebrain may account for the early appearance of apoptosis in response to hypoxIA as compared with that in hippocampus.
In vivo transfection of cis element “decoy” against nuclear factor- κB binding site prevents myocardial infarction
TLDR
The first successful in vivo transfer of NFκB decoy oligodeoxynucleotides to reduce the extent of myocardial infarction following reperfusion is reported, providing a new therapeutic strategy for myocardia infarctions.
Activation of the Nuclear Factor-κB Is a Key Event in Brain Tolerance
TLDR
Evidence is provided that activation of NFκB is a crucial step in the signal transduction pathway that underlies the development of brain tolerance and may open new strategies in the prevention of cerebral diseases, such as ischemia or epilepsy.
Transcription factor decoy for NFκB inhibits TNF-α-induced cytokine and adhesion molecule expression in vivo
TLDR
The present data showed that NFκB decoy, but not scrambled, ODN abolished TNF-α induced gene expression in vivo, as well as glomerular inflammation as assessed by CD45 staining, and suggest the potential utility of NFκBs decoy strategy for molecular therapy toglomerular inflammatory diseases.

The authors' results demonstrate that nuclear factor-κB decoy oligodeoxynucleotides transfected by cationic liposome method inhibited tumor necrosis factor —induced expression of interleukin-6 and intracellular adhesion molecule-1 in endothelial cells.
Lack of the p50 Subunit of Nuclear Factor-κB Increases the Vulnerability of Hippocampal Neurons to Excitotoxic Injury
TLDR
An important role for the p50 subunit of NF-κB in protecting neurons against excitotoxic cell death is demonstrated, as well as enhanced elevations of intracellular calcium levels and increased levels of oxidative stress after exposure to glutamate.
