Since the finding of activating mutation of BRAF in cutaneous melanoma we have learned to appreciate the remarkable diversity of the molecular mechanisms used by melanoma cells and the critical role played by the MAPK pathway in melanocyte and melanoma biology (Davies et al., 2002). The key role of MAPK in melanoma biology has been substantiated by using independent strategies for inhibiting RAS-RAF-MEK-ERK signaling.
