}Ribavirin (1-beta-D-ribofuranosyl-1,2,4-triazole-3-carboxamide) at concentrations of 10 microgram/ml or more, inhibited the replication of infectious pancreatic necrosis virus (IPNV) in both Chinook salmon embryo (CHSE-214) and rainbow trout gonad (RTG-2) cells. The drug was most effective when added just before or within 8 h p.i. Incorporation studies with radioactive precursors demonstrated that ribavirin suppressed cellular DNA and RNA synthesis within 2 to 3 h after addition of the drug…

Ribavirin is an antiviral that could be used to prevent ISAV replication either in vitro or in vivo, and in vivo assays showed a reduction in the mortality of Salmo salar by more than 90% in fish infected with ISAV and treated with ribavirin without adverse effects.

Eleven effective compounds against IHNV infection in vitro and in vivo were selected which showed the 50% inhibition of CPE spot within 10 μg/ml and were not observed the cytotoxicity within 10μm; some of the mode of action of compounds will be discussed.

The hypothesis that IHNV is less efficient at completing its growth cycle in cells with a simultaneous infection with IPNV is supported and can be affected at several stages of viral infection and is more sensitive to the action of antiviral compounds.

Results suggest that the antiviral activity of Virazole might be due to the inhibition of GMP biosynthesis in the infected cell at the step involving the conversion of IMP to xanthosine 5'-phosphate, which would result in inhibition of the synthesis of vital viral nucleic acid.
