Objective:We studied the possible interaction between itraconazole, a potent inhibitor of CYP3A, and zopiclone, a short-acting hypnotic. Methods:A double-blind, randomized, two-phase crossover design was used. Ten healthy young subjects received daily either 200 mg itraconazole or placebo for 4 days. On day 4 they ingested a single 7.5-mg oral dose of zopiclone. Plasma concentrations of zopiclone and itraconazole were determined and pharmacodynamic responses were measured up to 17 h.Results… Expand
Effect of itraconazole on the pharmacokinetics and pharmacodynamics of zolpidem
TLDR
The pharmacokinetics and pharmacodynamics of zolpidem were not remarkably affected by itraconazole in healthy volunteers, therefore, unlike triazolam, for example, zol pidem can be used in normal or nearly normal doses together with itraconsazole and probably also with other CYP3A4 inhibitors. Expand
Effect of itraconazole on the pharmacokinetics and pharmacodynamics of a single oral dose of brotizolam.
TLDR
Itraconazole increases plasma concentrations of brotizolam probably via its inhibitory effect on CYP3A4 brotzolam metabolism and the item 'sleepiness' of UKU were significantly decreased. Expand
No Effect of Itraconazole on the Single Oral Dose Pharmacokinetics and Pharmacodynamics of Estazolam
TLDR
The current study showed no significant effect of itraconazole on the single oral dose pharmacokinetics and pharmacodynamics of estazolam, suggesting that cytochrome P450 3A4 is not involved in the metabolism of estazerolam to a major extent. Expand
The CYP2C8 inhibitor gemfibrozil does not increase the plasma concentrations of zopiclone
TLDR
The possible effect of gemfibrozil, an inhibitor of CYP2C8, on the pharmacokinetics and pharmacodynamics of zopiclone was studied in vitro in human liver microsomes. Expand
Zopiclone: some remarks on pharmacokinetics and pharmacodynamics.
TLDR
In conclusion, adverse effects of zopiclone may occur under single-dose conditions or at steady-state, and the pharmacodynamic consequences may or may not closely follow pharmacokinetic changes. Expand
Pharmacokinetic evaluation of eszopiclone: clinical and therapeutic implications
TLDR
Overall, eszopiclone provides a therapeutic option for patients with sleep maintenance problems, though with accompanying potential for residual morning sedation, as well as a relatively high dollar cost of treatment. Expand
Possible Interaction of Zopiclone and Nefazodone
TLDR
The features observed in this case suggest the possibility of a drug—drug interaction between nefazodone and zopiclone, possibly resulting in enhanced hypnosedative efficacy in an 86-year-old white woman treated for depression and introduced for the management of insomnia. Expand
Comparative Pharmacokinetics and Pharmacodynamics of Short-Acting Hypnosedatives
  • D. Drover
  • Medicine
  • Clinical pharmacokinetics
  • 2004
TLDR
While zaleplon may be best indicated for the delayed onset of sleep, zolpidem and zopiclone may be better indicated for maintaining a complete night’s sleep. Expand
Zopiclone. An update of its pharmacology, clinical efficacy and tolerability in the treatment of insomnia.
TLDR
Although the available data on rebound insomnia and dependence liability are encouraging, potential differences between zopiclone and the benzodiazepines in these respects may have little clinical relevance in the context of short term intermittent use of hypnotics, as it currently recommended. Expand
Clinically Important Drug Interactions with Zopiclone, Zolpidem and Zaleplon
TLDR
Few clinically important interactions appear to have been reported in the literature with zaleplon, zolpidem and zopiclone, and the fact that these drugs are newer to the market and have not been as extensively studied as the conventional benzodiazepines may be the reason for this. Expand

The effect of erythromycin on the pharmacokinetics and pharmacodynamics of zopiclone.
TLDR
The interaction between erythromycin and zopiclone resulted mainly in accelerated absorption which may lead to a faster hypnotic effect in patients. Expand
Midazolam should be avoided in patients receiving the systemic antimycotics ketoconazole or itraconazole
Interaction between ketoconazole, itraconazole, and midazolam was investigated in a double‐blind, randomized crossover study of three phases at intervals of 4 weeks. Nine volunteers were given eitherExpand
Zopiclone. A review of its pharmacodynamic and pharmacokinetic properties and therapeutic efficacy as an hypnotic.
TLDR
Zopiclone is a useful alternative to other hypnotics, especially in patients intolerant of residual effects the morning after taking an hypnotic, which has a short half-life of about 5 hours and no long acting metabolites. Expand
Pharmacokinetics and metabolism of zopiclone.
TLDR
In human volunteers, plasma half-life of ZD increases with age, while patients with liver or renal impairments show little or no modification of pharmacokinetic parameters. Expand
A potentially hazardous interaction between erythromycin and midazolam.
TLDR
Metabolism of both erythromycin and midazolam by the same cytochrome P450IIIA isozyme may explain the observed pharmacokinetic interaction. Expand
A pharmacokinetic drug interaction between erythromycin and triazolam.
TLDR
In accordance with previously reported drug interactions involving macrolide antibiotics, erythromycin appears to significantly inhibit the metabolism of triazolam. Expand
Midazolam kinetics
TLDR
The effect‐kinetics of the new benzodiazepine midazolam was evaluated in six subjects after single oral and intravenous doses and infusion programs, and good intraindividual linear correlations were found between plasma levels and dynamic effects. Expand
Caffeine moderately antagonizes the effects of triazolam and zopiclone on the psychomotor performance of healthy subjects.
TLDR
It is concluded that triazolam may not differ importantly from diazepam as regards their antagonism towards caffeine, whereas further research on the antagonism between zopiclone and caffeine needs to be done. Expand
Drug-alcohol interactions on psychomotor skills: zopiclone and flunitrazepam.
TLDR
The results suggest that flunitrazepam, unlike zopiclone, has residual effects and interacts with alcohol in the morning following overnight ingestion of the drug. Expand
Itraconazole drastically increases plasma concentrations of lovastatin and lovastatin acid
TLDR
Itraconazole is a potent inhibitor of CYP3A4, and a possible interaction between these drugs was studied to study the risk of skeletal muscle toxicity following conjunction use of these drugs. Expand
