We studied the influence of metabolites of γ-aminobutyric shunt of the tricarboxylic acid cycle on the activities of aconitate hydratase (EC.4.2.1.3) as well as NAD- and NADP-specific isocitrate dehydrogenases (EC.1.1.1.41 and EC.1.1.1.42, respectively) using purified enzyme preparations from pea leaves (Pisum sativum L.) and maize scutellum (Zea mays L.). γ-Aminobutyric acid and succinate proved to have no significant effect on these enzymes, while 0.1–0.2 mM glutamate markedly activated NADP… Expand

Decrease in Manganese Superoxide Dismutase Leads to Reduced Root Growth and Affects Tricarboxylic Acid Cycle Flux and Mitochondrial Redox Homeostasis1[C][W]
TLDR
Transgenic Arabidopsis plants were generated in which expression of AtMSD1, encoding the mitochondrial manganese (Mn)SOD, was suppressed by antisense, demonstrating that reduced MnSOD affects mitochondrial redox balance and plant growth. Expand

Cloning and Expression Analysis of the Cytosolic NADP+-Dependent Isocitrate Dehydrogenase from Potato (Implications for Nitrogen Metabolism)
TLDR
In leaves, enzyme activities were dependent on the age, with fully mature leaves showing the highest level of RNA expression and enzyme activity, which may indicate that NADP+-dependent ICDH is not only involved in amino acid biosynthesis via the glutamine synthetase/glutamine oxoglutarate aminotransferase cycle but also in cycling, redistribution, and export of amino acids. On the function of the NADP-dependent isocitrate dehydrogenase isoenzymes in living organisms
TLDR
This review makes the point on the current knowledge of the physiological role of the E. coli, yeast and mammalian enzymes with the higher plant ICDH isoenzymes and the convenience of utilizing these data to study the physiological roles of the plant I CDHs is discussed. Expand
Are isocitrate dehydrogenases and 2-oxoglutarate involved in the regulation of glutamate synthesis?
TLDR
Several observations indicate that 2-oxoglutarate is likely to be a metabolic signal that regulates the coordination of carbon:nitrogen metabolism, discussed in the context of recent advances in bacterial signalling processes. Expand
Citrate and isocitrate in plant metabolism.
TLDR
The routes for citrate and isocitrate conversions are incorporated into the system of cross-linked metabolic processes and may provide carbon skeletons for nitrogen assimilation and reducing equivalents for biosynthesis, support the functioning of the glyoxylate cycle and play an important role in the TCA and energy metabolism as a whole. Publisher Summary The chapter discusses the preparation of aconitase enzyme, which is a simplification of the alcohol fractionation method of Morrison, followed by column chromatography. In freshExpand
The Tricarboxylic Acid Cycle in Plant Mitochondria: Its Operation and Regulation
A major role of plant mitochondria is to generate usable energy required for cellular processes. This is achieved by oxidizing organic acids in the Krebs or tricarboxylic acid (TCA) cycle (Fig. 1)Expand
Metabolism, enzymology and possible roles of 4-aminobutyrate in higher plants.
TLDR
Increase in 4-aminobutyrate content and glutamate decarboxylase activity have been observed in many plants under a variety of environmental stress conditions. Expand
Proton/l-Glutamate Symport and the Regulation of Intracellular pH in Isolated Mesophyll Cells.
TLDR
The data indicate that (a) continued H(+)/l-glutamate contransport is dependent upon GAD activity, (b) the pH-dependent properties of GAD are consistent with a role in a metabolic pH-stat, and (c) the regulation of intracellular pH during H( +)/ l-Glu symport may involve both H(+) consumption during 4-aminobutyrate production and ATP-driven H(+, efflux. Kinetic mechanism of potato tuber succinate semialdehyde dehydrogenase
TLDR
The results of these studies were consistent with an ordered bi, bi sequential mechanism for the potato enzyme in which NAD+ was the first substrate to bind to the enzyme and NADH was the last product to dissociate from it. Expand
