Different mediators released by anaphylaxis seem to be involved in different pathophysiological conditions, including cardiac arrhythmia. Histamine, 5-HT and platelet-activating factor (PAF) could participate in the enhanced arrhythmogenicity during anaphylaxis in guinea-pigs. The threshold dose of ouabain-induced arrhythmia is decreased in actively sensitized guinea-pigs by i.p. administration of ovalbumin. The purpose of the present paper was to investigate the effect of different mediator… Expand

Relevance of mediators to cardiac parameters of isolated anaphylactic guinea-pig heart.
TLDR
The data suggest that different mediators are time-dependently involved in anaphylaxis-induced cardiac changes, and cimetidine inhibited the tachycardia and clemastine reduced bradyarrhythmia. Expand
63. A New Perspective for Identifying Potential Cardiac Sensitizers
TLDR
The study demonstrates that several mathematical models can be constructed to predict the onset of ouabain induced arrhythmia, in both the dog and the swine, and that there are similarities in the two animal models. Expand
POTASSIUM CHANNEL OPENERS AND OTHER REGULATORS OF KATP CHANNELS
1. Interest in ATP‐sensitive K (Katp) channels first arose when it was shown that hypoglycaemic sulphonylureas, such as glibenclamide, closed these channels in pancreatic p‐cells to cause insulinExpand
Detecting Drug Promiscuity Using Gaussian Ensemble Screening
TLDR
The results indicate that GES is a useful way to study polypharmacology relationships, and it could provide a novel way to propose new targets for drug repositioning. Expand

TLDR
The results suggest that PAF could play a role in cardiac arrhythmias under special conditions and it supports the hypothesis thatPAF action is mediated by the release of leukotrienes and thromboxane. Expand
The arrhythmogenic effect of platelet activating factor (PAF) is inhibited by PAF antagonist and by substances influencing eicosanoids.

The present study supports the hypothesis that PAF aggravates cardiac arrhythmias under pathophysiological conditions in which PAF is involved. Expand

Effect of BN 52021, a specific PAF-acether antagonist, on cardiac anaphylaxis in Langendorff hearts isolated from passively sensitized guinea-pigs.
TLDR
Findings suggest that PAF-acether plays a major role as mediator in cardiac anaphylaxis, and BN 52021 may be a valuable therapeutic agent in allergic conditions. Expand
Ginkgolide B protects isolated hearts against arrhythmias induced by ischemia but not reperfusion.
TLDR
The antiarrhythmic effect of BN 52021 appears to be related to an antagonism of an increase in slow calcium influx induced by PAF in myocardial cells, which can presumably prevent the re-entry mechanism involved in the development of ischemia-induced rhythm disturbances. Expand

TLDR
The results suggest that the release of endogenous PGE2, PGF2α and prostacyclin could reduce early post‐infarction ventricular arrhythmias whilst the protective effect of aspirin in this model adds further support for the hypothesis that thromboxane release is involved in the genesis of these arrh rhythmias. Acetyl glyceryl ether phosphorylcholine (AGEPC). A putative mediator of cardiac anaphylaxis in the guinea pig.
TLDR
The results of these studies document that, during anaphylaxis in the isolated guinea pig heart, a platelet-activating factor is released into the coronary effluent that has physicochemical and functional properties similar to those of acetyl glyceryl ether phosphorylcholine. Expand
