Pentosidine is a fluorescent protein cross-link and glycoxidation marker for the advanced glycation reaction in diabetes, aging, and uremia. We raised polyclonal antibodies in New Zealand White rabbits against this hapten coupled to keyhole limpet hemocyanin. The antibodies detected by ELISA reacted strongly with free pentosidine but not with pentosidine-like compounds. The working range of the competitive ELISA for standard pentosidine was 0.1-100 pmol. Pentosidine was detectable in bovine… Expand
Methods of determination of pentosidine – the advanced glycation end-product of proteins
Glycation is a multi-step process that occurs spontaneously without the presence of enzyme, leading to the formation of AGEs – Advanced Glycation End-products. Glycation occurs in living organisms asExpand
The pentosidine concentration in human blood specimens is affected by heating
TLDR
The increase in pentosidine formation was significantly inhibited by acidic conditions more than by the addition of diethylene triamine pentaacetic acid, a metal chelator, which indicates that the level of serumpentosidine will be measured more accurately by ELISA if hydrochloric acid is added during the heating process. Formation of Pentosidine Cross-Linking in Myoglobin by Glyoxal: Detection of Fluorescent Advanced Glycation End Product
TLDR
In vitro glycation reaction of glyoxal on monomeric heme protein myoglobin results in AGE-induced protein modifications, which might be associated with complications in disease conditions. Expand
Measurement of pentosidine in biological samples.
TLDR
Pentosidine is a highly fluorescent advanced glycation end product (AGE) and crosslink derived from one molecule of arginine and one of lysine bridged in an imidazo-pyridinium structure that has now been detected and quantified in a variety of human and animal tissues. Expand

The serum concentration of the advanced glycation end-product Ne-(carboxymethyl)lysine is increased in uremia. Advanced glycation end products (AGEs) such as pentosidine and Ne-(carboxymethyl)lysineExpand
Plasma levels of pentosidine in diabetic patients: an advanced glycation end product.
TLDR
It is demonstrated that plasma pentosidine level was significantly influenced by the quality of glycemic control and renal function, and might be taken as a biomarker of diabetic cardiovascular risk. Expand

The first antibodies capable of recognizing each of the three isomers of the methylglyoxal hydroimidazolones (MG-Hs) by using chemical synthesis to create homogenous immunogens are generated. Expand
Plasma Pentosidine Levels Measured by a Newly Developed Method Using ELISA in Patients with Chronic Renal Failure
TLDR
The quantitative measurement method developed is judged to be a superior innovation for measuring pentosidine in body fluids and may be useful for the early diagnosis of mild renal failure and to estimate the degree of the severity of renal diseases. Expand
Direct quantification of pentosidine in urine and serum by HPLC with column switching.
TLDR
Using HPLC with column switching, a direct method of measuring pentosidine in urine and serum is developed that does not require pretreatment of samples and is convenient and useful for measuring urinary and serumpentosidine. Expand

Chromatographic Quantitation of Plasma and Erythrocyte Pentosidine in Diabetic and Uremic Subjects
TLDR
Plasma pentosidine may be a useful marker for monitoring the biochemical efficacy of trials with aminoguanidine or other treatment modalities and may act as a signal for advanced glycosylation end product-mediated receptor uptake by macrophages and other cells and contribute to accelerated atherosclerosis in diabetes and uremia. Expand
Immunohistochemical detection of advanced glycosylation end products in diabetic tissues using monoclonal antibody to pyrraline.
TLDR
Highly specific pyrraline immunoreactivity was detected in sclerosed glomeruli from diabetic and old normal kidneys as well as in renal arteries with arteriolosclerosis and in perivascular and peritubular sclerose extracellular matrix and basement membranes, which strengthens the notion that the advanced glycosylation reaction may contribute to decreased turnover and thickening of the extacellular matrix in diabetes and aging. Expand
Immunochemical approach to characterize advanced glycation end products of the Maillard reaction. Evidence for the presence of a common structure.
TLDR
The results strongly suggest the presence of a common structure in AGE preparations, regardless of whether AGE products are generated from proteins, amino acids, or monoaminocarboxylic acids. Expand
Immunochemical detection of advanced glycosylation end products in vivo.
TLDR
It is suggested that tissue AGEs which form in vivo appear to contain a common immunological epitope which cross-reacts with A GEs prepared in vitro, supporting the concept that immunologically similar AGE structures form from the incubation of sugars with different proteins. Expand
Aging of proteins: immunological detection of a glucose-derived pyrrole formed during maillard reaction in vivo.
TLDR
The pathobiological relevance of pyrraline may relate to its reported antiproteolytic and mutagenic properties and may play a role in diabetic neuropathy in analogy to pyrroles formed during hexane-induced neuropathy. Expand
End-stage renal disease and diabetes catalyze the formation of a pentose-derived crosslink from aging human collagen.
TLDR
Pentosidine emerges as a useful tool for assessment of previously unrecognized disorders of pentose metabolism in aging and disease and might be used as a measure of integrated pentosemia in analogy to glycohemoglobin for the assessment of cumulative glycemia. Expand
Pentosidine Formation in Skin Correlates With Severity of Complications in Individuals With Long-Standing IDDM
TLDR
A high correlation betweenpentosidine levels and long-wave collagen-linked fluorescence also was observed, suggesting that pentosidine is a generalized marker of accelerated tissue modification by the advanced glycosylation/Maillard reaction, which is enhanced in IDDM patients with severe complications. These findings provide the first immunochemical evidence that human lens crystallins contain advanced glycation end products and that these products increase with tissue age. Expand
Mechanism of formation of the Maillard protein cross-link pentosidine. Glucose, fructose, and ascorbate as pentosidine precursors.

The discovery that pentosidine can form not only from pentoses but also from hexoses and ascorbate raises major new questions concerning biochemical pathways of the Maillard reaction in vivo. Expand
Maillard Reaction-Mediated Molecular Damage to Extracellular Matrix and Other Tissue Proteins in Diabetes, Aging, and Uremia
TLDR
Immunohistochemical localization studies revealed that pyrraline is found predominantly in the sclerosed extracellular matrix of glomerular and arteriolar renal tissues from both diabetic and aged nondiabetic individuals, suggesting that molecular damage by advanced Maillard reaction products may be a common mechanism in their development.
