Water soluble 2'-taxol poly(ethylene glycol) (PEG) esters have been synthesized and shown to function in vitro as prodrugs. However, in vivo experiments clearly establish that in order for these prodrugs to behave in a predictable fashion, the molecular weight of PEG must be of such magnitude so as to maintain a t1/2(circulation) > t1/2(hydrolysis). When PEG derivatives of molecular weight approximately 40 kDa were employed with paclitaxel, ca. 4% by weight of paclitaxel was carried by the…

Synthesis and evaluation of water-soluble poly(vinyl alcohol)-paclitaxel conjugate as a macromolecular prodrug.
TLDR
It was suggested that the water-solubility of PTX was markedly enhanced by the conjugation to PVA, and PVA-SPTX effectively delivered PTX to the tumorous tissue due to the enhanced permeability and retention (EPR) effect.
Drug delivery of anticancer agents: water soluble 4-poly (ethylene glycol) derivatives of the lignan, podophyllotoxin.
Poly (ethylene glycol) Prodrugs: Altered Pharmacokinetics and Pharmacodynamics
TLDR
The exploitation of PEG prodrugs has been especially effective with anticancer agents, but other drugs in the areas of antibacterials and antifungals have also been successfully modified with PEG to produce pro drugs that demonstrate the desired increased AUCs.
A Polymeric Prodrug of 5-Fluorouracil-1-Acetic Acid Using a Multi-Hydroxyl Polyethylene Glycol Derivative as the Drug Carrier
TLDR
This study provides a new approach for the development of the delivery to tumors of anticancer agents with PEG derivatives, and possesses an exceedingly high 5-FA loading efficiency, much higher than the maximum drug loading efficiency of unmodified PEG with the same molecular weight.
Nano-sized assemblies of a PEG-docetaxel conjugate as a formulation strategy for docetaxel.
TLDR
The anti-cancer activity of the drug in the PEG-DTX micelle formulation was demonstrated to be retained in three human cancer cell lines and the maximum tolerated dose of this DTX formulation was also 2.5-fold higher than that for Taxotere(R) in healthy mice.
Effective drug delivery by PEGylated drug conjugates.
Synthesis and anti-cancer efficacy of rapid hydrolysed water-soluble paclitaxel pro-drugs
TLDR
The pro-drug was potent to inhibit the growth of various cancer cell lines, such as human lung, ovarian, colon and melanoma cancer cells and on the development of melanoma lung colonies in C57B/6 mice following intravenous administration of metastatic murine B16/F10 melanoma cells, the pro- drug seems to be more efficacious than paclitaxel.
A Biodegradable Polymeric Carrier Based on PEG for Drug Delivery
TLDR
A biodegradable copolymer, based on poly(ethylene glycol) (PEG) and fumarate (fum) units, was synthesized and studied as a drug carrier, overcoming the limitations of linear PEGs.
Preparation, characterization, cytotoxicity and pharmacokinetics of liposomes containing water-soluble prodrugs of paclitaxel.
Synthesis and In Vitro Evaluation of Polyethylene Glycol-Paclitaxel Conjugates for Lung Cancer Therapy
TLDR
Two different polyethylene glycol-paclitaxel ester conjugates with molecular weights of 6 and 20 kDa have the potential to retain pac litaxel in the lung for a prolonged duration and to sustain its release locally for a better efficacy.

Synthesis of congeners and prodrugs. 3. Water-soluble prodrugs of taxol with potent antitumor activity.
TLDR
Observations suggested that esters at C-2' and/or C-7, which would tend to promote water solubility, might serve as useful prodrugs of taxol, and salts had improved antitumor activity as compared to the free acids.
Distribution and tissue uptake of poly(ethylene glycol) with different molecular weights after intravenous administration to mice.
TLDR
The results of pharmacokinetic analysis suggested that small PEG tended to freely translocate from the circulation to extravascular tissues and to return to the blood circulation again by diffusion, whereas large PEG translocated more slowly to extraVascular tissues.
Polymer Conjugates
TLDR
More clinical pharmacokinetics studies are urgently needed to permit the validation of appropriate pharmacokinetic models that can be used in the future to assist in the optimisation of clinical protocols, and improved conjugate design.

Design, synthesis and biological activity of protaxols
TLDR
In vitro these prodrugs have cytotoxic properties against tumour cell lines comparable to those of taxol; moreover, human plasma catalyses the release of active taxol, and have greater potential as anticancer agents than the parent compounds taxol and taxotere.
Synthesis and characterization of poly(ethylene glycol) derivatives
TLDR
Five general routes for the preparation of polyoxyethylene derivatives are described and potential applications to catalysis, cell purifications, and other areas are discussed briefly.
