SummaryThe quinolone antibacterials are prone to many interactions with other drugs. Quinolone absorption is markedly reduced with antacids containing aluminium, magnesium and/or calcium and therapeutic failure may result.Other metallic ion-containing drugs, such as sucralfate, iron salts, and zinc salts, can also reduce absorption. Some of the newer quinolones inhibit the cytochrome P450 system, e.g. enoxacin, pefloxacin and ciprofloxacin. The toxicity of drugs that are metabolised by the…
Fluoroquinolones: Interaction Profile during Enteral Absorption
TLDR
This paper is a review of drug interactions with different quinolone derivatives at the absorption phase; the review deals mainly with the prototype qu inolones, ciprofloxacin and ofloxacins, and also with some of the newer agents.
Pharmacokinetic-Pharmacodynamic Drug Interactions with Nonsteroidal Anti-Inflammatory Drugs
TLDR
Gastrointestinal bleeding caused by NSAIDs is the most dangerous when it results from a mixed pharmacokinetic/pharmacodynamic interaction; however, patients are also at risk when pharmacodynamic interactions only are involved.
Pharmacokinetic Aspects of Treating Infections in the Intensive Care Unit
TLDR
Hydrophilic anti-infective agents are often eliminated unchanged by renal glomerular filtration and tubular secretion, and are therefore involved in competition for excretion, and there is a risk of nephrotoxicity and neurotoxicity with some cephalosporins and carbapenems.
Alteration of pharmacokinetics of cyclophosphamide and suppression of the cytochrome P450 genes by ciprofloxacin
TLDR
It is revealed that ciprofloxacin interacts with CY and suppressed relevant cytochromes P450 at the transcriptional level and this study may have a great clinical impact when ciproprofl oxacin is used in therapy.
Circumventing Drug Interactions
TLDR
The ability to prevent or minimize adverse drug interactions is of vital importance in optimizing the appropriate and effective use of antimicrobials and enhancing patient outcome.
An Overview of Fluoroquinolones
TLDR
An overview of fluoroquinolones, their mechanism of action, their uses, resistance development and their adverse effects is given.
Drug interaction and pharmacological considerations during anti-infective therapy in cancer patients.
TLDR
The purpose of this review is to indicate the drug-drug interactions that have true clinical relevance and may be found throughout the entire pharmacokinetic process (oral absorption, tissue distribution, excretion).
Drug Interactions with Cisapride
TLDR
A review of the published literature on the pharmacokinetic and pharmacodynamic interactions of cisapride with concomitantly administered drugs, providing clinicians with practical recommendations for avoiding these potentially fatal events.

TLDR
Drug interactions with the quinolones are of two types: pharmacokinetic and pharmacodynamic; there is evidence that the elderly and patients with liver disease are particularly susceptible to kinetic interactions with ciprofloxacin, and there is no evidence to suggest that ofloxacIn is likely to impair hepatic drug elimination.
Drug interactions of ciprofloxacin with other non-antibiotic agents.
TLDR
Ciprofloxacin and other quinolones inhibit gamma-aminobutyric acid receptors through reduction of their binding capacity and thus may potentiate convulsions induced by other agents, and cimetidine, a cytochrome P450 antagonist, affects metabolizable qu inolones, and it is likely that it may affect ciproFloxacIn's disposition after prolonged use.
Inhibition of Drug Metabolism by Quinolone Antibiotics
TLDR
Clinically, clinicians should be aware of the possibility of reduced drug clearance resulting in adverse effects whenever the fluoroquinolones are coadministered with drugs that depend on hepatic metabolism for their elimination, in combination with theophylline.

TLDR
The pharmacodynamic and pharmacokinetic drug interactions and pharmaceutical compatibilities of fluoroquinolones are reviewed and it is recommended to use non-interacting quinolones or to measure theophylline levels and reduce caffeine intake where appropriate.
Update on quinolone drug interactions.
Effect of a new quinolone, sparfloxacin, on the pharmacokinetics of theophylline in asthmatic patients
TLDR
Findings indicate that a once-daily dose of 200 mg of sparfloxacin has no significant effect on the pharmacokinetics and metabolism of theophylline and that it would be safe to coadminister this quinolone to asthmatic patients receiving chronic theophyLLine therapy.
Influence of oral co-administered metallic drugs on ofloxacin pharmacokinetics.
TLDR
Substantial correlation between in-vitro and in- vivo availability data was demonstrated in all cases except for ofloxacin combinations with ferrous sulphate and calcium carbonate, and in general, there was also good correlation between the saliva and urine data.
