Plasmids comprising transgene insertions in four lines of transgenic mice have been retrieved by plasmid rescue into a set of Escherichia coli strains with mutations in different members of the methylation-dependent restriction system (MDRS). Statistical analysis of plasmid rescue frequencies has revealed that the MDRS loci detect differential modifications of the transgene insertions among mouse lines that show distinctive patterns of transgene expression. Plasmids in mice that express hybrid… Expand
Other transgenic mutation assays: A new transgenic mouse mutagenesis test system using Spi− and 6‐thioguanine selections
TLDR
The results suggest that the new transgenic mouse “gpt delta” could be useful for the efficient detection of point mutations and deletion mutations in vivo. Expand
Identification and characterization of a gene responsible for inhibiting propagation of methylated DNA sequences in mcrA mcrB1 Escherichia coli strains
TLDR
The data presented here suggest that this locus is mrr and that the mrr gene product can recognize and restrict cytosine-methylated sequences and allow a significant improvement in the efficiency and representation of eukaryotic genomic and cDNA libraries. Expand

TLDR
A simple procedure for modifying BACs through homologous recombination using a targeting construct containing properly situated Chi sites is reported, demonstrating less mosaic and improved GFP expression in hematopoietic progenitor cells compared with smaller plasmid constructs. Expand
Expression Cloning of Recombinant Escherichia coli lacZ Genes Encoding Cytoplasmic and Nuclear β-galactosidase Variants
TLDR
The expression of cytoplasmic and nuclear variant of LacZ gene confirmed the ability of pcDNA3.1 as versatility nonviral vector for the experimental gene delivery study in mammalian cells. Expand
DNA modification and functional delivery into human cells using Escherichia coli DH10B.
TLDR
A novel comprehensive Escherichia coli-based vector system for the modification, propagation and delivery of large human genomic BAC clones into mammalian cells, using BAC host strain E.coli DH10B will greatly improve the utility of the available BAC libraries from the human and other genomes for gene expression and functional genomic studies. Direct Cloning of Isogenic Murine DNA in Yeast and Relevance of Isogenicity for Targeting in Embryonic Stem Cells
TLDR
A novel method for the direct cloning of genomic DNA into a targeting vector, pRTVIR, using oligonucleotide-directed homologous recombination in yeast is described, demonstrating the applicability of the method by constructing functional targeting vectors for mammalian genes Uhrf1 and Gfap. Expand
DNA modi ® cation and functional delivery into human cells using Escherichia coli DH 10 B
The availability of almost the complete human genome as cloned BAC libraries represents a valuable resource for functional genomic analysis, which, however, has been somewhat limited by the abilityExpand
Rapid and efficient cloning of proviral flanking fragments by kanamycin resistance gene complementation.
TLDR
This work cloned numerous proviral flanking fragments from growth factor-independent cell mutants that were obtained after infection with a replication incompetent retroviral vector and identified integrations into the cyclin D2 and several unknown genomic sequences. Expand
ENU-induced mutagenesis at a single A: T base pair in transgenic mice containing phi X174.
TLDR
Responses of a single A:T base pair to alkylation-induced mutation in a nonexpressed gene are demonstrated and the role of cell proliferation in somatic mutagenesis in vivo in higher eukaryotes is demonstrated. Expand
A Novel Selection Marker for Efficient DNA Cloning and Recombineering in E. coli
TLDR
It is shown that mfabI is not only an efficient plasmid selection marker, but it also possesses unique activity that may facilitate molecular manipulation of unstable sequences and expand the limited repertoire of selection markers in E. coli. Expand
