Cell and tissue cultures of Catharanthus roseus have been studied extensively as an alternative strategy to improve the production of valuable secondary metabolites. The purpose of this study was to produce C. roseus callus and suspension cell biomass of good quality and quantity to improve the total alkaloids and bis-indole alkaloids. The young stem derived-callus of C. roseus variety Quang Ninh (QN) was grown on MS medium supplemented with 1.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) plus…

The results showed when the pH rises, the concentration of MDA increases as the activities of antioxidative enzymes (peroxidase and catalase) rises, and the critical enzymes of alkaloid synthesis includephenylalanine ammonialyase and tryptophan decarboxylase in suspension cells were also found to increase, along with the alkaloid yield rises.

The results of qRT-PCR indicated that the increasing accumulations of vindoline, catharanthine, and ajmaline in C. roseus CMCs were correlated with the increasing expression of the above genes, which indicated that A. flavus fungal elicitor could enhance the TIA production of C.roseus C MCs, which might be used as an alternative biotechnological resource for obtaining bioactive alkaloids.
