In this study, the morphology, cytology and crossability of amphidiploid hybrids (2n = 8x = 80; WWW′W′TTT′T′) between Nicotiana wuttkei × Nicotianatabacum were studied. An R1 progeny of the hybrids were produced through in vitro chromosome doubling of the N. wuttkei × N. tabacum cv. Wiślica amphihaploids. The resulting amphidiploids showed enough self-fertility to be perpetuated by self-pollination. The plants preserved their phenotypes and the number of chromosomes over subsequent R2–R4 selfed…

To induce chromosome doubling, the protocorm-like bodies of a given variety were treated in vitro for 6 to 48 h with 10 to 160 mg L−1 amiprofos-methyl (APM) and plantlets with a swollen and shorter stem were observed after the APM treatment and these plants were also chromosome-doubled and became a distinctive morphological marker for the primary selection of chromosome- doubled plants.
