The cyclopentenone prostaglandins PGA2, PGA1, and PGJ2 are formed by dehydration within the cyclopentane ring of PGE2, PGE1, and PGD2. PGJ2 is metabolized further to yield Δ12‐PGJ2 and 15‐deoxy‐Δ12,14‐PGJ2 (15d‐PGJ2). Various compounds within the cyclopentenone prostaglandin family possess potent anti‐inflammatory, anti‐neoplastic, and anti‐viral activity. Most actions of the cyclopentenone prostaglandins do not appear to be mediated by binding to G‐protein coupled prostanoid receptors. Rather… Expand
Effects of Cyclopentenone Prostaglandins on the Expression of Heme Oxygenase‐1 in MCF‐7 Cells
TLDR
The induction of HO‐1 expression and the activation of Akt/PKB by 15d‐PGJ2 and PGA2 are likely to confer cytoprotective or antiapoptotic effects exerted by these cyPGs. Expand
Modification of Proteins by Cyclopentenone Prostaglandins is Differentially Modulated by GSH in Vitro
TLDR
Results evidence the importance of GSH‐cyP and/or GSH–protein interactions for the selectivity of protein modification by cyP and suggest a complex role for GSH that may be related to its ability to prevent protein oxidation or induce conformational alterations. Expand
Cyclopentenone Isoprostanes Inhibit the Inflammatory Response in Macrophages*
TLDR
It is suggested that cyclopentenone isoprostanes may serve as negative feedback regulators of inflammation and have important implications for defining the role of oxidative stress in the inflammatory response. Expand
Molecular pharmacology of the human prostaglandin D2 receptor, CRTH2
TLDR
Functional studies demonstrated that PGD2 activation of recombinant CRTH2 results in decrease of intracellular cAMP in a pertussis toxin‐sensitive manner and showed that CRTH 2 can functionally couple to the G‐protein Gαi/o. Expand
Cyclopentenone Prostaglandins: Biologically Active Lipid Mediators Targeting Inflammation
TLDR
These properties render them unique therapeutic value, especially in resolving inflammation and could be used in adjunct with other existing therapies, as well as discuss other α, β -unsaturated carbonyl lipids and cyPGs like isoprostanes (IsoPs) compounds. Expand
Prostanoids with Cyclopentenone Structure as Tools for the Characterization of Electrophilic Lipid–Protein Interactomes
TLDR
A detailed knowledge of targets of electrophilic eicosanoids and the functional consequences of their modification will contribute to the understanding of their mechanism of action and help assess whether these endogenous mediators can be exploited as the basis for the development of novel therapeutic strategies. Expand
Effects of 15‐Deoxy‐Δ12,14‐prostaglandin J2 on the Expression of p53 in MCF‐7 Cells
TLDR
The electrophilic carbon center located in the α,β‐unsaturated carbonyl moiety of the cyclopentenone ring might be critical for the control of DNA‐binding activity as well as cellular levels of p53 by 15d‐PGJ2. Expand
Prostaglandins of J Series Control Heme Oxygenase Expression
TLDR
CyPGs appear to possess a promising future in designing therapeutics for many neurologic diseases, such as Alzheimer's disease, vascular‐related dementia, multiple sclerosis, ischemic conditions, and many others in which inflammation is a part of the pathophysiology. Expand
Proteomic identification of the candidate target proteins of 15‐deoxy‐delta12,14‐prostaglandin J2
TLDR
This protein data set highlights the potential for 15d‐PGJ2 to covalently modify cellular proteins and provides a source of data that will aid further studies on the mechanism of action of this endogenous regulator of inflammation. Expand
Cyclopentenone prostaglandins PGA2 and 15‐deoxy‐δ12,14 PGJ2 suppress activation of murine microglia and astrocytes: Implications for multiple sclerosis
TLDR
Comparisons of the relative effectiveness of 15d‐PGJ2 and PGA2 in inhibiting the inflammatory response of mouse microglia and astrocytes demonstrate for the first time that PGA 2 is a potent antiinflammatory mediator. Expand
