Membrane-associated TNF-alpha cleavage is required to yield the 17.5-kD soluble product. This process is poorly understood in human cells, and no studies have related this process to the alveolar macrophage (AM). TNF-alpha-converting enzyme (TACE) is known to cleave TNF at the Ala-76-Val-77 site. We have evaluated the expression, regulation, and catalytic function of TACE in healthy human AMs. TACE was detected on the surface of AMs using flow cytometry. TACE protein can be upregulated by LPS…

Studies on the Role of Acid Sphingomyelinase and Ceramide in the Regulation of Tumor Necrosis Factor α (TNFα)-converting Enzyme Activity and TNFα Secretion in Macrophages*
TLDR
This study shows that ASMase is a negative regulator of LPS-induced tumor necrosis factor α (TNFα) secretion in macrophages, and the underlying mechanism for these effects is not transcriptional but post-translational.
Investigation of the Role of TNF-α Converting Enzyme (TACE) in the Inhibition of Cell Surface and Soluble TNF-α Production by Acute Ethanol Exposure
Toll-like receptors (TLRs) play a fundamental role in the immune system by detecting pathogen associated molecular patterns (PAMPs) to sense host infection. Ethanol at doses relevant for humans
Tumour necrosis factor‐α processing in interstitial lung disease: a potential role for exogenous proteinase‐3
TLDR
Evidence is provided for PR‐3‐mediated cleavage in UIP, which may have implications for future therapeutic targeting of TACE, especially in diseases with a neutrophil component typical of usual interstitial pneumonia.
Release of soluble tumor necrosis factor receptor 1 from corneal epithelium by TNF-alpha-converting enzyme-dependent ectodomain shedding.
TLDR
TACE-dependent ectodomain shedding of sTNFR1 was recognized in corneal epithelium and may play an anti-inflammatory role in the inflammatory condition.
Reactive Oxygen Species and p38 Mitogen-activated Protein Kinase Mediate Tumor Necrosis Factor α-Converting Enzyme (TACE/ADAM-17) Activation in Primary Human Monocytes*
TLDR
In primary human monocytes, ROS mediate LPS-induced up-regulation of TACE activity indirectly through activation of the p38 signaling pathway, which redefines the mechanisms of Tace activation in primary cells with a physiological stimulus.
Studies of TNF-alpha in Alpha-One Antitrypsin Deficient and healthy subjects
TLDR
In monocytes from A1ATD-related COPD subjects, trends were observed for TNFR1 blockade to reduce both types of cytokine, for IL-10 to be reduced by TNFR2 blockade and forTNFR1 expression at the monocyte surface to be up-regulated by TNF-α-TNFR2 binding.
Signaling pathways involved in LPS induced TNFalpha production in human adipocytes
TLDR
This study clearly demonstrates that the LPS induced activation pathway is an integral part of the inflammatory process linked to obesity, and that adipocytes are responsible for most of the secreted TNFalpha in inflamed adipose tissue, through TLR4 activation.
Membrane versus soluble isoforms of TNF-α exert opposing effects on tumor growth and survival of tumor-associated myeloid cells.
TLDR
It is shown for the first time that membrane and soluble isoforms have diametrically opposing effects on both tumor growth and myeloid content and that there are significant differences in the role of different TNF-α isoforms in tumor progression and the bioavailability of each isoform may distinctly regulate tumor progression.

The fate of pro-TNF-alpha following inhibition of metalloprotease-dependent processing to soluble TNF-alpha in human monocytes.
TLDR
The addition of hydroxamic acid-based metalloprotease inhibitors to the cell culture is capable of blocking >95% of the production of soluble TNF-alpha and leads to a transient, but reproducible, increase in cell surface T NF-alpha as measured by FACS analysis.
Release of tumour necrosis factor alpha (TNFalpha) by TNFalpha cleaving enzyme (TACE) in response to septic stimuli in vitro.
TLDR
The regulation of memTNF by TACE on monocytes is an important regulatory event in the pro-inflammatory cytokine cascade and is suggested to play a role in the pathophysiology of sepsis.

The regulation of memTNF by TACE on monocytes is an important regulatory event in the pro-inflammatory cytokine cascade and may play a role in the pathophysiology of sepsis.
In Vitro Processing of Human Tumor Necrosis Factor-α (*)
TLDR
A potential role for this enzyme as an accessory TNF-α processing enzyme is suggested following detection of a serine proteinase processing activity present in crude membrane preparations of monocytic cells able to generate a 17-kDa active protein.
Functional analysis of the domain structure of tumor necrosis factor-alpha converting enzyme.
TLDR
It is found that TACE must be expressed with its membrane-anchoring domain for phorbol ester-stimulated shedding of TNF, p75 TNFR, and IL-1R-II, but that the cytoplasmic domain is not required for the shedding of these substrates.

It is shown that the release of mature TNF-alpha from leukocytes cultured in vitro is specifically prevented by synthetic hydroxamic acid-based metalloproteinase inhibitors, which also prevent the release into the circulation of endotoxin challenged rats.
