A recombinant of herpes simplex virus (HSV) was constructed in which the HSV thymidine kinase (TK) gene was deleted and the varicella-zoster virus (VZV) TK gene was introduced into the US5 region under the control of the human cytomegalovirus IE promoter. Infection with the recombinant (R18) led to the induction of TK although the kinetics of synthesis resembled those of a 'late' gene product. The recombinant was virulent in the zosteriform mouse model with the pattern of pathogenesis similar…

Differences in the Likelihood of Acyclovir Resistance-Associated Mutations in the Thymidine Kinase Genes of Herpes Simplex Virus 1 and Varicella-Zoster Virus
TLDR
The results suggest that the relatively low frequency of ACV-resistant VZV is a consequence of the characteristics of the TK gene.
Herpesvirus saimiri pathogenicity enhanced by thymidine kinase of herpes simplex virus.
TLDR
Recombinant viruses for the functional expression of the thymidine kinase (TK) of herpes simplex virus type 1 (HSV) as a suicide gene reliably allowed the targeted elimination of transduced nonpermissive human T cells in vitro after the administration of ganciclovir.
Crystal Structure of Varicella Zoster Virus Thymidine Kinase*
TLDR
The crystal structure of Varicella zoster virus (VZV, human herpes virus 3) thymidine kinase complexed with (E)-5-(2-bromovinyl)-2′-deoxyuridine 5′-monophosphate and ADP is determined and should be of use in the design of novel antiherpes and antitumor drugs.
Human Cytomegaloviruses Expressing Yellow Fluorescent Fusion Proteins - Characterization and Use in Antiviral Screening
TLDR
Three recombinant cytomegaloviruses carrying the enhanced yellow fluorescent protein (EYFP) fused with the viral proteins IE-2, ppUL32, and ppUL83 behaved all like wild type, even at low multiplicity of infection (MOI).
Recombinant Green Fluorescent Protein-Expressing Human Cytomegalovirus as a Tool for Screening Antiviral Agents
TLDR
The GFP-based antiviral assay appeared to be very reliable for measuring virus-inhibitory effects in concentration- and time-dependent fashions and might also be adaptable for high-throughput screenings of cytomegalovirus-specific antiviral agents.
Highly purified recombinant varicella Zoster virus thymidine kinase is a homodimer.
TLDR
Determinations of the native molecular weight by size exclusion chromatography and native polyacrylamide gel electrophoresis revealed that the pure enzyme is biologically active as a homodimer.
Molecular characterization and antiapoptotic function analysis of the duck plague virus Us5 gene
TLDR
The results show that the DPV Us5 gene is a late gene and that the Us5 protein is a component of the virion, is localized in the cytoplasm, and can inhibit apoptosis induced by H2O2 in DEF cells.

The role of herpes simplex virus type 1 thymidine kinase in pathogenesis.
TLDR
Inoculation of mice with the HSV TK deletion mutant resulted in the establishment of latent ganglionic infection as demonstrated by superinfection of explanted ganglia with wild-type (wt) virus but not by routine explant culture suggesting that the virus-encoded TK is not essential for theestablishment of latent infection but may be necessary for either reactivation or virus replication following reactivation.
Activity of the simian virus 40 early promoter-enhancer in herpes simplex virus type 1 vectors is dependent on its position, the infected cell type, and the presence of Vmw175
TLDR
Results show that while HSV gene products expressed in the presence of Vmw175 inhibited SV40-cat expression in the tk locus in a cell-type-specific manner, HSV genes expressed inThe presence or absence of V mw 175 inhibited SV 40-catexpression in the BamHI z locus independently of cell type.
Rapid identification of nonessential genes of herpes simplex virus type 1 by Tn5 mutagenesis.
TLDR
A simple insertional mutagenesis method has been used for selective inactivation of individual HSV-1 genes and three genes proved to be nonessential for herpes simplex virus replication in Vero cells and the US4 gene appeared to be involved in viral pathogenesis in the central nervous system of mice.
An analysis of the in vitro and in vivo phenotypes of mutants of herpes simplex virus type 1 lacking glycoproteins gG, gE, gI or the putative gJ.
TLDR
It is concluded that the gE-gI complex plays a part, at least in some cell types, in the interactions at the cell surface that allow transmission of the virus from infected to uninfected cells by cell contact.
Analysis of mutations in the thymidine kinase genes of drug-resistant varicella-zoster virus populations using the polymerase chain reaction.
TLDR
In vitro amplification using the PCR combined with cloning and sequencing is a relatively rapid method for identifying mutations in small virus populations even when they are not homogeneous.
Divergent molecular pathways of productive and latent infection with a virulent strain of herpes simplex virus type 1
TLDR
It is concluded that replication-competent HSV type 1 can establish latency without initiating productive infection.
In vitro susceptibility of varicella-zoster virus to acyclovir
TLDR
Analysis of the metabolism of acyclovir in varicella-zoster virus-infected WI-38 cells revealed that, as with herpes simplex virus types 1 and 2, the formation of the triphosphate forms of the drug is specific to viral infection.
Zosteriform spread of herpes simplex virus as a model of recrudescence and its use to investigate the role of immune cells in prevention of recurrent disease
TLDR
Observations indicate that recrudescent lesions appear in the presence of cell populations normally associated with rapid virus clearance; cellular immune mechanisms may be rendered ineffective owing to the lack of recruitment to the site of recrudescence until tissue breakdown instigates an inflammatory response.
Mechanism of selective inhibition of varicella zoster virus replication by 1-beta-D-arabinofuranosyl-E-5-(2-bromovinyl)uracil.
TLDR
It can be concluded that marked inhibition of VZV replication by BV-araU is due to selective phosphorylation of BV -araU in the TK+ VZ V-infected cells and strong inhibition ofVZV DNA synthesis by Bv-ara U triphosphate, without detectable incorporation into VZv DNA.
