Bacillus amyloliquefaciens NK-1 has the potential to produce levan and poly-gamma-glutamic acid (γ-PGA) simultaneously. However, it is not possible to purify each single product from the same strain because the extraction process is identical. We deleted the pgs cluster (for γ-PGA synthesis) from the NK-1 strain and constructed a γ-PGA-deficient NK-ΔLP strain. Nuclear magnetic results showed that the NK-ΔLP strain could produce high purity levan product. However, its levan titer was only 1.96 g…

Recruiting a new strategy to improve levan production in Bacillus amyloliquefaciens
TLDR
This is the first report of the improvement of levan production in microbes deficient inextracellular proteases and TasA, and the NK-Q-7 strain exhibits outstanding characteristics for extracellular protein production or extracllular protein related product synthesis.
Improvement of levan production in Bacillus amyloliquefaciens through metabolic optimization of regulatory elements
TLDR
This is the first report of the improvement of levan production through metabolic optimization for sacB expression and levansucrase secretion, and provided essential insights for systematically metabolic engineering of microbial cell factories for enhanced biochemical production.
Biosynthesis and prebiotic activity of a linear levan from a new Paenibacillus isolate.
TLDR
Microbiome and metabolome analysis showed that Plev intervention increased the abundance of beneficial bacteria and elevated the levels of short-chain fatty acids (SCFAs) in feces of mice, and could be considered a potential thickener and prebiotic supplement in food industry.
Efficient production of levan using a recombinant yeast Saccharomyces cerevisiae hypersecreting a bacterial levansucrase
TLDR
A cost-effective and industrially applicable production system for food-grade levan was developed using a Saccharomyces cerevisiae protein secretion system and recombinant yeast secreting RaLsrA could produce levan more efficiently by microbial fermentation.
Development of a cost-effective production process for Halomonas levan
TLDR
The development of a cost-effective production process was achieved with the highest yield reported so far on this microbial system, as well as the highest theoretical bioconversion efficiency ever reported for levan-producing suspension cultures.
Enhanced production of antifungal lipopeptide iturin A by Bacillus amyloliquefaciens LL3 through metabolic engineering and culture conditions optimization
TLDR
The first report on simultaneous production of four iturin A homologs (C14–C17) by a Bacillus strain is reported, suggesting that metabolic engineering in combination with culture conditions optimization may be a feasible method for enhanced production of bacterial secondary metabolites.
icrobial levan from Brachybacterium phenoliresistens : haracterization and enhancement of production arek
Levan producing bacteria was isolated from rhizosphere soil. The molecular identification of this isolate was conducted using 16S rRNA, which resulted in a sequenced region of 1298 base pairs. The
Engineering of a genome-reduced strain Bacillus amyloliquefaciens for enhancing surfactin production
TLDR
The genome-reduced strain GR167 was advantageous over the parental strain in several industrially relevant physiological traits assessed and it was highlighted as a chassis strain for further genetic modification.

selective production and characterization of levan by Bacillus subtilis (Natto) Takahashi.
TLDR
B. Takahashi strain is the most efficient levan-producing strain among all of the B. subtilis strains tested and it produced the highest yield of levan in the least time under the common cultivation condition.
Functions of poly-gamma-glutamic acid (γ-PGA) degradation genes in γ-PGA synthesis and cell morphology maintenance
TLDR
This is the first report concerning the markerless deletion of γ-PGA degradation genes to improve γ"-PGA production in a glutamate-independent strain and the first observation that γ -glutamyltranspeptidase (encoded by ggt) could be involved in the inhibition of cell elongation.

Biosynthesis and Hydrolysis of Poly(γ-glutamic acid) from Bacillus subtilis IF03335.
TLDR
Hydrolytic degradation of PGA was found to proceed through a random chain scission and a by-product was produced, which seemed to be a polysaccharide.
Curing the Plasmid pMC1 from the Poly (γ-glutamic Acid) Producing Bacillus amyloliquefaciens LL3 Strain Using Plasmid Incompatibility
TLDR
The analysis of the strain bearing incompatible plasmids after incubation at 30 °C for 30 generations showed the production of plasmid cured strains and high frequency of elimination was achieved with more than 93 % of detected strains showing to bePlasmid-cured.
Microbial production and chemical transformation of poly-γ-glutamate
TLDR
The occurrence of multiple carboxyl residues in PGA likely plays a role in its relative unsuitability for the development of bio‐nylon plastics and thus, establishment of an efficient PGA‐reforming strategy is of great importance.
