Acute myeloid leukemia (AML) is characterized by molecular heterogeneity. As commonly altered genomic regions point to candidate genes involved in leukemogenesis, we used microarray-based comparative genomic hybridization and single nucleotide polymorphism profiling data of 391 AML cases to further narrow down genomic regions of interest. Targeted resequencing of 1000 genes located in the critical regions was performed in a representative cohort of 50 AML samples comprising all major…

Epimutations mimic genomic mutations of DNMT3A in acute myeloid leukemia
TLDR
Aberrant DNA hypermethylation within the DNMT3A gene, in analogy to DN MT3A mutations, is frequently observed in AML and both modifications seem to be useful for risk stratification or choice of therapeutic regimen.
Epigenetic modifications of splicing factor genes in myelodysplastic syndromes and acute myeloid leukemia
TLDR
It is concluded that DNA hypermethylation does not frequently silence splicing factors in MDS and AML, however, in the case of YBX3, promoter hyper methylation‐induced downregulation may contribute to the pathogenesis or maintenance of AML.
Profiling of somatic mutations in acute myeloid leukemia with FLT3-ITD at diagnosis and relapse.
TLDR
This study clearly shows that FLT3-ITD AML requires additional driver genetic alterations in addition to FLT2-ITd alone, and DNMT3A mutations are the most stable mutations.
A Genome-Wide Aberrant RNA Splicing in Patients with Acute Myeloid Leukemia Identifies Novel Potential Disease Markers and Therapeutic Targets
TLDR
The results suggest that aberrant splicing is a common characteristic for AML and suggest that splice variant transcripts that are the result of splicing aberrations create novel disease markers and provide potential targets for small molecules or antibody therapeutics for this disease.
The modular network structure of the mutational landscape of Acute Myeloid Leukemia
TLDR
A single in silico model was defined and a set of seed-genes that may trigger leukemogenesis in patients with normal karyotype were established that suggested a specific alteration could not be as essential as the interaction of deregulated pathways.
Gene mutations of acute myeloid leukemia in the genome era
TLDR
This review focuses on gene mutations that were recently discovered by genome sequencing that apparently increase the complexity and heterogeneity of acute myeloid leukemia.
Driver mutations of cancer epigenomes
TLDR
Cancer epigenetics is a relatively new, promising frontier with much potential for improving cancer outcomes, and therapies such as 5-azacytidine and decitabine have proven that targeting epigenetic alterations in cancer can lead to tangible benefits.
The Mutational Landscape of Acute Promyelocytic Leukemia Reveals an Interacting Network of Co-Occurrences and Recurrent Mutations
TLDR
A network analysis of multiple mutations provides a reliable approach to prioritize genes for additional analysis, improving the knowledge of the leukemogenesis interactome and defined a functional module in the interactome of APL.
NOTCH2 and FLT3 gene mis-splicings are common events in patients with acute myeloid leukemia (AML): new potential targets in AML.
TLDR
The results suggest that NOTCH2 and FLT3 mis-splicing is a common characteristic of AML and has the potential to generate transcripts encoding proteins with altered function and splice variants of these genes might provide disease markers and targets for novel therapeutics.

Exome sequencing identifies somatic mutations of DNA methyltransferase gene DNMT3A in acute monocytic leukemia
TLDR
The identification of somatic mutations by exome sequencing in acute monocytic leukemia, the M5 subtype of acute myeloid leukemia (AML-M5), suggests a contribution of aberrant DNA methyltransferase activity to the pathogenesis of acute monocrytic leukemia and provides a useful new biomarker for relevant cases.

TLDR
A large spectrum of genomic imbalances, including novel recurring changes in AML with complex karyotypes, was identified using array-CGH, allowing high-resolution genome-wide screening of genomicImbalances.
Recurring mutations found by sequencing an acute myeloid leukemia genome.
TLDR
By comparing the sequences of tumor and skin genomes of a patient with AML-M1, recurring mutations that may be relevant for pathogenesis are identified.
Identification of acquired copy number alterations and uniparental disomies in cytogenetically normal acute myeloid leukemia using high-resolution single-nucleotide polymorphism analysis
TLDR
The data show the potential of high-resolution SNP analysis for identifying genomic regions of potential pathogenic and clinical relevance in AML, and there seemed to be an association between UPD 11p and UPD 13q cases with overall survival.
TP53 alterations in acute myeloid leukemia with complex karyotype correlate with specific copy number alterations, monosomal karyotype, and dismal outcome.
TLDR
TP53 alteration is the most important prognostic factor in CK-AML, outweighing all other variables, including the MK category, and in multivariable analysis for overall survival, TP53 alterations, white blood cell counts, and age were the only significant factors.
KIT mutations confer a distinct gene expression signature in core binding factor leukaemia
TLDR
The data suggest that this CBF leukaemia subgroup might profit from TKI therapy, however, the relevance of the KIT mutation‐associated signature remains to be validated prior to clinical implementation.
Frequent mutation of histone modifying genes in non-Hodgkin lymphoma
TLDR
Genes with roles in histone modification were frequent targets of somatic mutation and suggest a previously unappreciated disruption of chromatin biology in lymphomagenesis in NHLs.
The Genetic Landscape of the Childhood Cancer Medulloblastoma
TLDR
Key differences between the genetic landscapes of adult and childhood cancers are demonstrated, dysregulation of developmental pathways as an important mechanism underlying MBs is highlighted, and a role for a specific type of histone methylation in human tumorigenesis is identified.

TLDR
AML with RUNX1 mutations are characterized by distinct genetic properties and are associated with resistance to therapy and inferior outcome, and Explorative subgroup analysis revealed that allogeneic hematopoietic stem-cell transplantation had a favorable impact on RFS in RunX1-mutated patients.
CREBBP mutations in relapsed acute lymphoblastic leukaemia
TLDR
Analysis of an extended cohort of 71 diagnosis–relapse cases and 270 acute leukaemia cases that did not relapse found that 18.3% of relapse cases had sequence or deletion mutations of CREBBP, which encodes the transcriptional coactivator and histone acetyltransferase CREB-binding protein (CREBBP).
