In drug discovery today, drug exposure is determined in preclinical efficacy and safety studies and drug effects are related to measured concentrations rather than to the administered dose. This leads to a strong increase in the number of bioanalytical samples, demanding the development of higher throughput methods to cope with the increased workload. Here, a combined approach is described for the high-throughput preparation and liquid chromatography/tandem mass spectrometry (LC/MS/MS) analysis…
Development of a novel high-throughput analytical methodology, multiple injection method, for quantitative analysis in drug metabolism and pharmacokinetic studies using liquid chromatography with tandem mass spectrometry.
The peak intensity was found to be unaffected by overlapping compounds even if they have wide range of ionization efficiency, and no difference was shown in the quantification values of MIM compared to the conventional methods.

Internal standard tracked dilution to overcome challenges in dried blood spots and robotic sample preparation for liquid chromatography/tandem mass spectrometry assays.
An alternative sample dilution for liquid chromatography/tandem mass spectrometry (LC/MS/MS) assays using IS to track the dilution step is introduced as "IS-tracked dilution", which was proven to be effective and convenient in both plasma assays and DBS Assays using omeprazole as a probe compound.
Practical use of the regression offset approach for the prediction of in vivo intrinsic clearance from hepatocytes
Three AstraZeneca projects are used to exemplify how a two-sided prediction interval, applied to the rat regression corrected reference data, can form the basis for assessing the likelihood that, for a given chemical series, the in vitro kinetic data is predictive of in vivo clearance and is therefore appropriate to guide optimisation of compound metabolic stability.
The volume of distribution is an indicator of poor in vitro–in vivo extrapolation of clearance for acidic drugs in the rat
It is shown that accurate IVIVE for acidic compounds with high plasma protein binding and low renal CL can be associated with a high Vd and with a low in vivo CL, and that Mw is an important optimization parameter for pharmacokinetic.

'It is better to be useful than perfect'. This review attempts to critically cover and assess the currently available approaches and tools to answer the crucial question: Is it possible (and if it

Cassette-accelerated rapid rat screen: a systematic procedure for the dosing and liquid chromatography/atmospheric pressure ionization tandem mass spectrometric analysis of new chemical entities as part of new drug discovery.
Using this new approach, a threefold increase in throughput was achieved over the previously reported "rapid rat screen", and the liquid chromatography/atmospheric pressure ionization tandem mass spectrometry (LC/API-MS/MS) assay is also streamlined by analyzing the samples as "cassettes of six".
