The cyclic undecapeptide cyclosporin A (CyA), one of the most valuable immunosuppressive drugs, is produced nonribosomally by a multifunctional cyclosporin synthetase enzyme complex by the filamentous fungus Tolypocladium niveum. To increase CyA productivity by wild-type T. niveum (ATCC 34921), random mutagenesis was first performed using an antifungal agar-plug colony assay (APCA) selection approach. This generated a mutant strain producing more than 9-fold greater CyA than the wild-type…

Recent advances in understanding the structure, function, and biotechnological usefulness of the hemoglobin from the bacterium Vitreoscilla
The hemoglobin from the bacterium Vitreoscilla (VHb) is the first microbial hemoglobin that was conclusively identified as such (in 1986). It has been extensively studied with respect to its ligand

TLDR
Several ActinomyCetes/Streptomycetes expression vectors are described for expression of the Vitreoscilla haemoglobin gene (vhb) in an industrial erythromycin-producing strain of Saccharopolyspora ERYthraea, showing that vhb is expressed in an active form.

Identification of TmcN as a pathway-specific positive regulator of tautomycetin biosynthesis in Streptomyces sp. CK4412.
TLDR
It is suggested that TmcN is a pathway-specific positive regulator that activates transcription of the TMC biosynthetic pathway genes in Streptomyces sp.
Heterologous Expression of Novel Cytochrome P450 Hydroxylase Genes from Sebekia benihana
TLDR
Six novel CYP genes isolated from a rare actinomycetes called Sebekia benihana were functionally expressed in Streptomyces lividans and showed strong hydroxylation activities toward 7-ethoxycoumarin, suggesting potential application of act inomycETes CYPs for the biocatalysts of natural product bioconversion.
Expression of vitreoscilla hemoglobin improves growth and levels of extracellular enzyme in yarrowia lipolytica
TLDR
The expression of VHb in this yeast was found to enhance growth, contrary to reported observations in wild‐type Saccharomyces cerevisiae in which there was no significant growth enhancement.

TLDR
Two genes, dps encoding decaprenyl diphosphate synthase and dxs encoding 1-deoxy-D-xylulose 5-phosphate synthesis, were isolated from Rhizobium radiobacter ATCC 4718 and carried out heterologous expression in Escherichia coli in order to identify their functions.

Vitreoscilla Hemoglobin
TLDR
The obligate aerobic bacterium, Vitreoscilla, synthesizes elevated quantities of a homodimeric hemoglobin under hypoxic growth conditions and has a cytoplasmic (not periplasmic) localization and is concentrated near the periphery of the cytosolic face of the cell membrane.
