Coloradocin was isolated from a fermentation broth by adsorption onto Amberlite XAD-2. The activity was eluted in MeOH and purified by gel filtration on Shephadex LH-20, followed by liquid-liquid chromatography on diol-bonded silica gel. The last two steps in the purification of this antibiotic included reverse-phase chromatography on C18-bonded silica gel and countercurrent chromatography on an Ito Coil Planet Centrifuge to give material of 90% purity. Analytically pure material was obtained… Expand

Isolation and purification of secondary metabolites.
TLDR
This chapter discusses the isolation and purification of secondary metabolites, which are often produced in very low yields by the original wild type strains and are no longer an activity for which there is much support in academic or industrial laboratories. Expand

Detection of novel secondary metabolites.
TLDR
This review focuses on the genesis and development of the gamut of methodologies that have led to the successful detection of the wide variety of novel secondary metabolites that include antibacterial, antigungal, antiviral and antitumour antibiotics, enzyme inhibitors, pharmacologically and immunologically active agents, products useful in agriculture and animal husbandry, microbial regulators, and other compounds for which no bioactive role has yet been found. The nargenicin family of oxa-bridged macrolide antibiotics.
TLDR
The history of this family of compounds is reviewed, including synthetic approaches, and the recently uncovered genetic basis for nargenicin production is highlighted, and structure-activity relationships among these molecules are investigated with a view to the future development of members of this unusual antibiotic family. TLDR
Three oxidoreductases that fix the required 10/6/6-tryciclic core are identified and crystal structure of the heterodimeric complex NtfO1-NtfO2, homologues of StmO 1-StmO2 with equivalent function, reveals protein-protein interactions. Expand
Toward the total synthesis of luminamicin; an anaerobic antibiotic: construction of highly functionalized cis-decalin containing a bridged ether moiety
TLDR
One of the essential steps in this synthetic route is construction of a cis-decaline framework using a one-pot Michael addition-aldol reaction of a conjugated aldehyde. Expand
