}In the present work we studied the karyotype stability during long-term in vitro maintenance in 3 cloned strains of Leishmania (Viannia) peruviana, Leishmania (Viannia) braziliensis and a hybrid between both species. Only the L. (V.) peruviana strain showed an unstable karyotype, even after subcloning. Four chromosomes were studied in detail, each of them characterized by homologous chromosomes of different size (heteromorphy). Variations in chromosome patterns during in vitro maintenance were…

Analysis of hybrids generated within and between different strains of L. tropica revealed genome wide patterns of recombination, demonstrating that classical crossing over occurs at meiosis, and allowed us to construct the first physical and genetic maps in Leishmania.
Haplotype selection as an adaptive mechanism in the protozoan pathogen Leishmania donovani
- BiologyNature Ecology & Evolution
- 2017
It is shown that aneuploidy turnover and haplotype selection are two mechanisms by which L. donovani adapts to environmental fluctuations inside the mammalian host and that karyotypic fluctuations allow for selection of beneficial haplotypes.
Stress conditions promote the mating competency of Leishmania promastigotes in vitro marked by expression of the ancestral gamete fusogen HAP2
- Biology

This work reveals that there are specific populations involved in Leishmania mating associated with a discernible transcriptomic signature, and that stress facilitated in vitro hybridization can be a transformative approach to generate large numbers of hybrid genotypes between diverse species and strains.

A substantial level of diversity was observed when compared with the reference genome, with SNP counts ranging from ~95,000 to ~131,000 for the different isolates, and both Z72 and Z75 isolates are characterized by a much reduced heterozygosity, consistent with the existence of distinct evolutionary groups in the restricted area sampled and a substantial genetic diversity within L. braziliensis.
