To clarify the clinical features of adult patients with acute leukemia (AL) with 11q23 abnormalities, we performed a retrospective analysis of data from 58 adult Japanese patients: 51 with acute myeloid leukemia (AML), and 7 with acute lymphoblastic leukemia (ALL). The incidences according to fusion partners in AML were: t(9;11), 31.3%; t(11;19), 27.4%; t(6;11), 21.5%. The incidence of patients with t(11;19) was higher than those in the US and Europe, and the incidence of t(4;11) was lower than… Expand
Prognostic significance of 11q23 aberrations in adult acute myeloid leukemia and the role of allogeneic stem cell transplantation
TLDR
Using intention-to treat analysis based on donor availability, it was noted that allogeneic stem cell transplant in first remission had a significant benefit toward improving OS and relapse-free survival in patients with AML and 11q23 abnormalities. Expand
Rearrangement of 11q23 Chromosome Region in Acute Myeloid Leukemias in Children
Aim. To study characteristics of 11q23 involvement, agespecific differences in the incidence of these chromosomal markers in acute myeloid leukemias (AML) in children, and to determine theirExpand
Allogeneic hematopoietic cell transplantation in adult acute myeloid leukemia with 11q23 abnormality: a retrospective study of the Adult Acute Myeloid Leukemia Working Group of the Japan Society for Hematopoietic Cell Transplantation (JSHCT)
TLDR
It is suggested that allo-HCT offered a curative option for adult AML with 11q23 abnormality and the prognostic value of minimal residual disease (MRD) status at HCT on transplant outcomes among hematological CR patients was the significant prognostic indicator for relapse and survival in CR patients. TLDR
Special strategies are needed to treat 11q23/MLL AL, including allogeneic hematopoietic stem cell transplantation, according to the fusion partner, and the development of novel methodologies are also needed to improve the prognosis. Expand

TLDR
Transplant age (>45 years), allo-HSCT in non-CR state adn positive minimal residual disease (MRD) before transplantation was a negative prognostic factor on DFS, confirming the higher TRM in patients at non- CR state when transplanted. Expand
Cytogenetic abnormalities in adult non-promyelocytic acute myeloid leukemia: a concise review.
TLDR
This review performs a concise description of more recurrent cytogenetic aberrations found in AML, their correlations with biological and clinical data and theirs strong impact with outcome of patients, useful for therapeutic decision. Expand
Rearrangement of the Myeloid/Lymphoid Leukemia Gene in Therapy-Related Myelodysplastic Syndrome in Patients Previously Treated with Agents Targeting DNA Topoisomerase II
TLDR
It is proposed that the MLL gene is etiopathogenetically relevant for hematological neoplasias transformation and survival and is associated with a worse overall survival. Expand
Efficacy of myeloablative allogeneic hematopoietic stem cell transplantation in adult patients with MLL-ELL-positive acute myeloid leukemia
TLDR
Intriguingly, clonally expanded non-leukemic cells expressing MLL-ELL during consolidation therapy were found to be eradicated after allo-HSCT during the monitoring of minimal residual disease in one patient; this indicates that allo -HSCT is efficacious for eliminating pre-leukmic cells resistant to chemotherapy. TLDR
Multivariate analysis showed that transplantation, platelets >50×109/L at onset, and CR are associated with a better OS in MLL rearranged AML patients, and extramedullary involvement is an independent prognostic factor for DFS. Expand
Genetic, Hematologic and Psychological Aspects of Leukemia
Current advances in diagnostic methods, our understanding of its molecular basis and therapeutics have made leukemia one of the most exciting and rapidly changing fields in oncology. Owing to theExpand

Adult patients with de novo acute myeloid leukemia and t(9;11)(p22;q23) have a superior outcome to patients with other translocations involving band 11q23: A Cancer and Leukemia Group B Study
TLDR
It is concluded that the outcome of adults with de novo AML and t(9; 11) is more favorable than that of Adults with other 11q23 translocations; this is especially true for t( 9;11) patients who receive intensive postremission therapy. Expand

TLDR
Within the subgroup of young adult AML patients with normal karyotype, MLL PTD is associated with short remission duration, and Multivariate analysis identified the MLLPTD status as the single prognostic factor for remission duration. Expand
Hematological malignancies with t(9;11)(p21–22;q23) – a laboratory and clinical study of 125 cases
TLDR
Among the t(9;11)+ve patients with AML, the white cell count (WBC) and age group were significant predictors of event-free survival; central nervous system (CNS) involvement or karyotype class (sole, with trisomy 8, or with other), also contributed to prognosis although the data could not show these to be independent factors. Expand
The t(6;11)(q27;q23) translocation in acute leukemia: a laboratory and clinical study of 30 cases
TLDR
The median overall survival for the patients was 12 months, indicating a poor prognosis for patients with a t(6;11) translocation, and molecular investigations showed that one case had a 3′ deletion of MLL. Expand
Rearrangement of ALL1 (MLL) in acute myeloid leukemia with normal cytogenetics.
TLDR
The partial tandem duplication of ALL1 was responsible for ALL1 rearrangement in all such cases examined, making it a frequent molecular defect in adult AML patients with normal cytogenetics and thus require new therapeutic approaches. Expand
Biology and treatment of infant leukemias.
TLDR
Rare congenital forms of lymphoid or myeloid leukemia, manifested at birth or during the first month of life, carry a dismal prognosis, especially when a MLL/11q23 rearrangement is present; such cases should be carefully distinguished by chromosomal/molecular analysis and cell culture techniques from transient myeloproliferative disorders. Expand
AML with 11q23/MLL abnormalities as defined by the WHO classification: incidence, partner chromosomes, FAB subtype, age distribution, and prognostic impact in an unselected series of 1897 cytogenetically analyzed AML cases.
TLDR
The category AML with 11q23/MLL abnormalities accounts for 2.8% of unselected AML, is closely associated with monocytic differentiation, and has a dismal prognosis. Expand
Chromosome abnormalities and MLL rearrangements in acute myeloid leukemia of infants
TLDR
This study showed that while 11q23/MLL rearrangements were also common in infant AML, AML infants with such rearrangement had a clinical outcome similar to that of AMl infants without such rearrANGements. Expand
Rearrangements of the MLL gene in therapy-related acute myeloid leukemia in patients previously treated with agents targeting DNA- topoisomerase II
TLDR
The association of exposure to topo II- reactive chemotherapy with 11q23 rearrangements involving the MLL gene in t-AML suggests thatTopo II may play a role in the aberrant recombination events that occur in this region both in AML de novo and in t -AML. Expand
