It has been reported that clarithromycin (CAM) augments the anti-tumor activity of thalidomide against multiple myeloma (MM) cells, while the mechanism remains unclear. A myeloma cell line or primary myeloma cells were treated with CAM. Autophagy was analyzed by morphological changes, LC3 expression and lysotracker staining. CAM induced vacuoles in the cytoplasm of MM cells which resembled autolysosomes. The manifestation of the CAM-induced vacuoles was blocked by treatment with PI3-kinase… Expand
A new purpose for an old drug: inhibiting autophagy with clarithromycin
TLDR
It is suggested that maneuvers to inhibit autophagy may restore disease sensitivity in patients with CML that has become refractory to the available tyrosine kinase inhibitors. Expand
Clarithromycin enhances bortezomib-induced cytotoxicity via endoplasmic reticulum stress-mediated CHOP (GADD153) induction and autophagy in breast cancer cells
TLDR
Simultaneously targeting two major intracellular protein degradation pathways such as the ubiquitin-proteasome system by BZ and the autophagy-lysosome pathway by CAM may improve the therapeutic outcome in breast cancer patients via ER-stress mediated apoptosis. Expand
Clarithromycin effectively enhances doxorubicin-induced cytotoxicity and apoptosis in MCF7 cells through dysregulation of autophagy.
PURPOSE Use of autophagy inhibitors in combination with chemotherapy has become a novel chemotherapeutic strategy. In this study, we aimed to determine whether the effectiveness of doxorubicin (DOX)Expand
Clarithromycin Synergistically Enhances Thalidomide Cytotoxicity in Myeloma Cells
TLDR
The interaction effects of CAM and thalidomide on myeloma cells are evaluated and data suggest that the efficacy of CAM against MM was partly due to its synergistic action with the immunomodulatory agents. Expand
Macrolides sensitize EGFR-TKI-induced non-apoptotic cell death via blocking autophagy flux in pancreatic cancer cell lines
TLDR
The possibility of using macrolides as ‘chemosensitizers’ for EGFR-TKI therapy in pancreatic cancer patients to enhance non-apoptotic tumor cell death induction is suggested. Expand
Macrolide antibiotics block autophagy flux and sensitize to bortezomib via endoplasmic reticulum stress-mediated CHOP induction in myeloma cells
TLDR
Simultaneously targeting two major intracellular protein degradation systems such as the ubiquitin-proteasome system by BZ and the autophagy-lysosomes system by a macrolide antibiotic enhances ER stress-mediated apoptosis in MM cells. Expand
Combined treatment with SAHA, bortezomib, and clarithromycin for concomitant targeting of aggresome formation and intracellular proteolytic pathways enhances ER stress-mediated cell death in breast cancer cells.
TLDR
The combined effect of vorinostat, which has a potent inhibitory effect for HDAC6, with CAM and BZ in breast cancer cell lines, and expression levels of ER-stress-related genes were maximally induced by the simultaneous combination of three reagents suggest that simultaneous targeting of intracellular proteolytic pathways andHDAC6 enhances ER- stress-mediated apoptosis in breast cancers. Expand
Host cell autophagy activated by antibiotics is required for their effective antimycobacterial drug action.
TLDR
It is demonstrated that isoniazid and pyrazinamide promote autophagy activation and phagosomal maturation in Mycobacterium tuberculosis (Mtb)-infected host cells and dampened Mtb-induced proinflammatory responses in macrophages, underscoring the importance of host autphagy in orchestrating successful antimicrobial responses to mycobacteria during chemotherapy. Expand
Clarithromycin inhibits autophagy in colorectal cancer by regulating the hERG1 potassium channel interaction with PI3K
TLDR
It is concluded that Cla affects the autophagic flux by impairing the signaling pathway linking hERG1 and PI3K, and combining Cla with 5-FU might be a novel therapeutic option in CRC. Expand

Radiation-induced autophagy is associated with LC3 and its inhibition sensitizes malignant glioma cells.
TLDR
It is shown that IR induced cell cycle arrest and autophagy, but not apoptosis, in human malignant glioma U373-MG cells, and Autophagy inhibitors, 3-methyladenine and bafilomycin A1, radiosensitized U373 -MG cells are shown to represent a new application of radiosensitization for malignantglioma cells. 3-Methyladenine suppresses cell migration and invasion of HT1080 fibrosarcoma cells through inhibiting phosphoinositide 3-kinases independently of autophagy inhibition.
TLDR
The results suggest that 3-MA suppresses the invasion of HT1080 cells, independently of autophagy inhibition, through inhibition of type I and II PI3Ks and possibly other molecules. Expand
Adjuvant Effect of Clarithromycin on Chemotherapy for Murine Lung Cancer
TLDR
It is indicated that CAM can exhibit an antitumor effect by itself and also induce the well-balanced expansion of helper T cell subsets in tumor-bearing mice recovering from the immunosuppression caused by chemotherapy. Expand
Clarithromycin is a potent inhibitor of tumor-induced angiogenesis

Clinithromycin is a potent inhibitor of tumor-induced angiogenesis that exerts its effect by inhibiting endothelial cell tube formation, and may be a possible candidate for therapeutic application. Expand

Autophagy—A double‐edged sword in oncology
TLDR
It is argued that further analyses are necessary before manipulation of autophagic mechanisms can find its way into tumour therapy for improved treatment of patients, and data demonstrating that cellular decisions to autophagy manipulation depend on tumour type, stage, microenvironment, Autophagy initiator or inhibitor and combined tumour treatment is discussed.
