Abstract. Objective: A possible pharmacokinetic interaction between a CYP1A2 inhibitor, ciprofloxacin, and clozapine was studied in schizophrenia patients with stable clozapine treatment. Methods: A randomised double-blind cross-over study design with two phases was used. Seven schizophrenic inpatients volunteered to receive, in addition to their previous drug regimen, either 250 mg ciprofloxacin or placebo twice daily (b.i.d.) for 7 days. The phases were separated by a 7-day wash-out period…

Ciprofloxacin and Clozapine: A Potentially Fatal but Underappreciated Interaction

Clinicians are advised to monitor baseline clozapine levels prior to adding strong CYP450 1A2 inhibitors, reduce the clozAPine dose by at least two-thirds if adding a 1A1 inhibitor such as ciprofloxacin, and adjust the clazapine dose to maintain levels close to those obtained at baseline.

Ciprofloxacin Strongly Inhibits Clozapine Metabolism

It is suggested that inhibition of cytochrome P450 (CYP) enzymes 1A2 and 3A4 by ciprofloxacin resulted in delayed clozapine metabolism and elevated clozAPine plasma concentrations, which might cause severe adverse effects.

Effect of ciprofloxacin on the pharmacokinetics of ropivacaine

Ciprofloxacin modestly decreased the mean ropivacaine CL by inhibiting the CYP1A2-mediated formation of 3-OH-ropivacane and the CYB3A4- mediated formation of PPX was increased.

A puzzling case of increased serum clozapine levels in a patient with inflammation and infection.

A 23-year old male with a history of schizophrenia treated with clozapine 900 mg/d was admitted to the hospital for a gastrointestinal infection. The trough serum concentration of clozapine at

Increase in plasma levels of clozapine after addition of isoniazid.

A report of how a possible pharmacokinetic drug interaction between clozapine (Clozaril) and isoniazid and a pharmacodynamic interaction between venlafaxine (Effexor) and has been predicted resulted in a non-harmful event that could have been fatal.

Interaction of clozapine and ciprofloxacin: a case report

A 47-year-old man has been an inpatient in a medium-security forensic psychiatry unit with a diagnosis of treatment-resistant paranoid schizophrenia and was advised that ciprofloxacin is the recommended treatment of acute epididymoorchitis caused most often by non-sexually-transmitted Gram negative enteric organisms in men older than 35 years.

Effect of caffeine-containing versus decaffeinated coffee on serum clozapine concentrations in hospitalised patients.

The effect of instant coffee drinking on serum clozapine concentrations is of minor clinical relevance in most of the patients, but some individuals may be more sensitive to this interaction due e.g. to genetic factors.

Effects of drug interactions and liver disease on the pharmacokinetics of ropivacaine

The results of the present studies demonstrate the principal role of hepatic CYP1A2, and suggest the modest role of CYP3A4, in the elimination of ropivacaine in humans in vivo.

Effect of ciprofloxacin and ibuprofen on thein vitro Metabolism of rosiglitazone and oral pharmacokinetics of rosiglitazone in healthy human volunteers

Both CFX and IBF did not affect the in vitro metabolism of RGZ in human liver microsomes and the pharmacokinetic parameters following co-administration of either CFX or IBF.
