Previous studies have revealed that nucleosomes impede elongation of RNA polymerase II (RNAPII). Recent observations suggest a role for ATP-dependent chromatin remodellers in modulating this process, but direct in vivo evidence for this is unknown. Here using fission yeast, we identify Fun30Fft3 as a chromatin remodeller, which localizes at transcribing regions to promote RNAPII transcription. Fun30Fft3 associates with RNAPII and collaborates with the histone chaperone, FACT, which facilitates…
It's fun to transcribe with Fun30: A model for nucleosome dynamics during RNA polymerase II-mediated elongation
TLDR
A model is presented that describes nucleosome dynamics during RNAPII-mediated elongation in mathematical terms and addresses the involvement of Fun30Fft3 in this process.
Structure and function of the chromatin remodeler SMARCAD1 with its nucleosome substrate
TLDR
It is shown that SMARCAD1 transfers the entire histoneOctamer from one DNA segment to another in an ATP-dependent manner but is also capable of de novo nucleosome assembly from histone octamer, due to its ability to bind all histones simultaneously.
The Mechanism of Chromatin Remodeler SMARCAD1/Fun30 in Response to DNA Damage
TLDR
The present review summarizes the recent advances in the functional and mechanistic studies on a particular chromatin remodeler SMARCAD1Fun30 which has been demonstrated to play a key role in distinct cellular processes and gained much attention in recent years.
Nucleosome Remodeling by Fun30SMARCAD1 in the DNA Damage Response
TLDR
The current knowledge of a chromatin remodeler Fun30SMARCAD1, which plays a key role in genome maintenance, is reviewed and a picture emerges in which commonalities within these context-dependent roles of Fun30 SMARCad1 exist, which may help to gain a more global understanding of chromatin alterations induced by Fun 30SMARC AD1.
Cockayne syndrome B protein acts as an ATP-dependent processivity factor that helps RNA polymerase II overcome nucleosome barriers
TLDR
It is shown that CSB forms a stable complex with Pol II and acts as an ATP-dependent processivity factor that helps Pol II across a nucleosome barrier, which is distinct from the canonical modes of chromatin remodelers that directly engage and remodel nucleosomes or transcription elongation factors that facilitate Pol IIucleosome bypass without hydrolyzing ATP.
Evidence that dissociation of Spt16 from transcribed genes is partially dependent on RNA Polymerase II termination
TLDR
Using the Saccharomyces cerevisiae system, evidence is provided that at least at some genes dissociation of the FACT subunit Spt16 from their 3′ ends is partially dependent on RNA Polymerase II (Pol II) termination.
Chromatin remodeler Fft3 plays a dual role at blocked DNA replication forks
TLDR
This work investigates the function of fission yeast Fun30/Smarcad1 family of SNF2 ATPase-dependent chromatin remodeling enzymes in DNA damage repair and finds that only Fft3 has a function in DNA repair and it is needed for single-Strand annealing of an induced double-strand break.
Structural visualization of key steps in nucleosome reorganization by human FACT
TLDR
The EM structure of the hexasome, generated by the addition of Mid-pAID or pAID, indicated that the dissociation of H2A-H2B dimer causes significant alteration from the canonical path of the nucleosomal DNA.

The nucleosomal barrier to promoter escape by RNA polymerase II is overcome by the chromatin remodeler Chd1
TLDR
It is found that Chd1 evicts nucleosomes downstream of the promoter in order to overcome the nucleosomal barrier and enable PolII promoter escape, thus providing mechanistic insight into the role of Chd 1 in transcription and pluripotency.
Spt6 Regulates Intragenic and Antisense Transcription, Nucleosome Positioning, and Histone Modifications Genome-Wide in Fission Yeast
ABSTRACT Spt6 is a highly conserved histone chaperone that interacts directly with both RNA polymerase II and histones to regulate gene expression. To gain a comprehensive understanding of the roles
Histone chaperone FACT action during transcription through chromatin by RNA polymerase II
TLDR
It is shown that human FACT (hFACT) facilitates transcription through chromatin and promotes nucleosome recovery in vitro and interacts with DNA-binding surfaces of H2A/H2B dimers, facilitating uncoiling of DNA from the histone octamer.
FACT Facilitates Transcription-Dependent Nucleosome Alteration
TLDR
The FACT facilitates Pol II–driven transcription by destabilizing nucleosomal structure so that one histone H2A-H2B dimer is removed during enzyme passage, defining the mechanism by which Pol II can transcribe through chromatin without disrupting its epigenetic status.
The FACT Complex Travels with Elongating RNA Polymerase II and Is Important for the Fidelity of Transcriptional Initiation In Vivo
TLDR
It is suggested that FACT contributes to the fidelity of Pol II transcription by linking the processes of initiation and elongation by ensuring that only appropriate mRNAs are synthesized.
