Remoxipride is an atypical antipsychotic displaying selective binding to the dopamine D2 receptor. Several cases of aplastic anemia led to the withdrawal of remoxipride from the market in December 1993. The remoxipride metabolite NCQ-344 is a hydroquinone while the structural isomer NCQ-436 is a catechol, both of which have been suggested to be capable of forming a reactive para- and ortho-quinone, respectively. Recently, these two remoxipride metabolites were shown to induce apoptosis in human… Expand
Quinoid derivatives of the nevirapine metabolites 2-hydroxy- and 3-hydroxy-nevirapine: activation pathway to amino acid adducts
Nevirapine (NVP) is the non-nucleoside HIV-1 reverse transcriptase inhibitor most commonly used in developing countries, both as a component of combined antiretroviral therapy and to preventExpand
Identification of the Thiophene Ring of Methapyrilene as a Novel Bioactivation-Dependent Hepatic Toxicophore
TLDR
A thiophene ring substituent is identified as a bioactivation-dependent toxicophore in hepatocytes for the first time and a possible causal linkage with cytotoxicity is established. Expand
IDENTIFICATION OF A NOVEL GLUTATHIONE ADDUCT OF DICLOFENAC, 4′-HYDROXY-2′-GLUTATHION-DESCHLORO-DICLOFENAC, UPON INCUBATION WITH HUMAN LIVER MICROSOMES
TLDR
The analysis of GS adducts from human liver microsome incubation with DF by the NanoMate 100-API 3000 method unambiguously revealed a new adduct ion, which was confirmed to be 4′-OH-2′-glutathion-deschloro-diclofenac by liquid chromatography tandem mass spectrometry (MS), LC/MS-NMR, and comparison to a synthetic standard. Expand
Characterization of glutathione conjugates of duloxetine by mass spectrometry and evaluation of in silico approaches to rationalize the site of conjugation for thiophene containing drugs.
TLDR
The in vitro bioactivation of the selective serotonin and norepinephrine reuptake inhibitor duloxetine was investigated and the utility of in silico approaches MetaSite and induced fit docking to address bioactivation liabilities was demonstrated. Expand
Differences in CYP3A4 catalyzed bioactivation of 5-aminooxindole and 5-aminobenzsultam scaffolds in proline-rich tyrosine kinase 2 (PYK2) inhibitors: retrospective analysis by CYP3A4 molecular docking, quantum chemical calculations and glutathione adduct detection using linear ion trap/orbitrap mass
TLDR
A retrospective analysis on representative 5-aminooxindole and 5-aminobenzsultam PYK-2 inhibitors utilizing CYP3A4 molecular docking and quantum chemical calculations revealed key differences in active site binding and two-electron oxidation rates, which correlate with GSH adduct formation with the two moieties. Synthesis and cyclic voltammetry studies of 3,4-methylenedioxymethamphetamine (MDMA) human metabolites
3,4-Methylenedioxymethamphetamine (MDMA or “Ecstasy”) is a widely abused, psychoactive recreational drug. There are growing evidences that the MDMA neurotoxic profile may be highly dependent on itsExpand
Molecular structure and dynamic properties of a sulfonamide derivative of glutathione that is produced under conditions of oxidative stress by hypochlorous acid.
TLDR
It is demonstrated that the GSH+2O-2H species is indeed a nine-membered cyclic sulfonamide, and elucidation of the structures of the glutathione sulfonamides provides further impetus to explore their potential as biomarkers of hypochlorous acid formation. Expand
Production of glutathione sulfonamide and dehydroglutathione from GSH by myeloperoxidase-derived oxidants and detection using a novel LC-MS/MS method.
TLDR
It is concluded that glutathione sulfonamide is sufficiently selective for HOCl to be useful as a biomarker for myeloperoxidase activity in biological systems and a novel oxidation product of GSH with a molecular weight two mass units less than GSH is named dehydroglutathione. Expand
2-Thioxanthines Are Mechanism-based Inactivators of Myeloperoxidase That Block Oxidative Stress during Inflammation*
TLDR
2-thioxanthines inhibited MPO in plasma and decreased protein chlorination in a mouse model of peritonitis and slowed but did not prevent neutrophils from killing bacteria and were poor inhibitors of thyroid peroxidase. Expand
Minimising the potential for metabolic activation in drug discovery
TLDR
Major themes explored in this review include a comprehensive cataloguing of bioactivation pathways of functional groups commonly utilised in drug design efforts with appropriate strategies towards detection of corresponding reactive intermediates. Expand
