}The peripheral mechanisms responsible for pressor response produced by microinjections of baclofen (GABA(B) agonist) into the nucleus tractus solitarii (NTS) of conscious rats were studied. Bilateral microinjections of baclofen (10-1,000 pmol/100 nl) produced a dose-related increase in mean arterial pressure (MAP) and heart rate. The maximal response was observed after 15 min. Intravenous injection of prazosin decreased MAP to control levels. Subsequent treatment with Manning compound…

The specificities of the baclofen-induced effects are probably related to peculiarities ofThe functioning of GABAB receptors, the activation of which may mediate the effects of multiple neuronal mechanisms.

Results indicate that chronic ANG II infusion stimulates GABA( B) receptor expression and augments GABA(B) receptor-mediated responses in the NTS, which could contribute to the central nervous system actions of ANG II that result in dampening of baroreflexes and elevation in arterial BP.

It is found that passive and voluntary cycling evokes similar increases in heart rate and that these responses were enhanced after diazepam administration, a benzodiazepine that enhances GABAA activity, suggesting that the GABAergic system may contribute to the muscle mechanoreflex-mediated vagal withdrawal at the onset of exercise in humans.

It is concluded that baclofen causes elevation of blood pressure in the nucleus tractus solitarii of conscious rats, and that this structure is a possible locus of action for systemically administered bacl ofen in producing hypertension.

The roles of vasopressin, the sympathoadrenal system, and the renin-angiotensin system in the production of hypertension after bilateral destruction of the nucleus tractus solitarius (NTS) were examined in chloralose-anesthetized rats and results suggest that this form of hypertension is due to increased sympathoadenal activity and increased vasoppressin release into plasma and that the ren in-angiotsin system is not involved.
