The dendritic cells (DC) of mouse spleen and thymus were examined for expression of CD4 and CD8. Provided care was taken to avoid selective extraction or selective depletion of DC subpopulations, three main types of DC were detected in mouse spleen: a major new population of CD4+8− DEC-205low CD11bhigh DC, together with the previously described CD4−8− DEC-205low CD11bhigh DC and CD4−8αα+ DEC-205high CD11blow DC. The CD4 on the surface of the CD4+ splenic DC subpopulation was produced by the DC…
Concept of lymphoid versus myeloid dendritic cell lineages revisited: both CD8α− and CD8α+dendritic cells are generated from CD4lowlymphoid-committed precursors
TLDR
Experiments of DC reconstitution after irradiation revealed that bothCD8α+ DCs and CD8α− DCs were generated after transfer of CD4low precursors, suggesting that both CD8β+ and CD7α−DCs derive from a common precursor and do not support the concept of the CD8 α+ lymphoid-derived and CD6α−myeloid- derived DC lineages.
Anatomic location and T-cell stimulatory functions of mouse dendritic cell subsets defined by CD4 and CD8 expression.
TLDR
Interestingly, both CD8alpha+ DCs and CD4-/CD8- DCs efficiently primed minor histocompatibility (H-Y male antigen) cytotoxicity following intravenous injection, whereas CD4+DCs were weak inducers of CTLs.
Spleen-Resident CD4+ and CD4− CD8α− Dendritic Cell Subsets Differ in Their Ability to Prime Invariant Natural Killer T Lymphocytes
TLDR
It is shown that, when loaded in vitro with OVA peptide or whole protein, and in steady-state conditions, splenic CD4− and CD4+ cDC are equivalent in their capacity to prime and directCD4+ and CD8+ T cell differentiation.
Identification of CD8alpha+CD11c- lineage phenotype-negative cells in the spleen as committed precursor of CD8alpha+ dendritic cells.
TLDR
It is reported here that murine splenic splenic CD8alpha+CD11c- lineage phenotype (Lin)- cells could differentiate into CD8 alpha+ DCs in vivo after intravenous transplantation, andFlow cytometry analysis and Immunohistochemistry staining suggest that CD8 Alpha+CD 11c-Lin- cells are a committed precursor ofCD8alpha- DCs.
Human thymus contains 2 distinct dendritic cell populations.
TLDR
WhereasCD11b+ thymic DCs appear to be related to tonsillar germinal-center DCs, the major CD11b- IL-12-secreting human thymus DC population has similarities to mouse CD 11b- CD8+ DCs.
Distinct functional capacities of mouse thymic and splenic dendritic cell populations
TLDR
The results indicate that the microenvironment is an important factor that contributes to the functional specification of DC subsets in different lymphoid tissues.
Identification of a CD8alpha(+) dendritic cell subpopulation in rat spleen and evaluation of its OX-62 expression.
TLDR
A CD8alpha(+) DC subpopulation in rat spleen less effective in inducing an immune response than CD8 alpha(-) DC is identified and the presence of DC subpopulations with different lineages in DC preparations based on OX-62 expression is suggested.
Concept of lymphoid versus myeloid dendritic cell lineages revisited : both CD 8 a 2 and CD 8 a 1 dendritic cells are generated from CD 4 low lymphoid-committed precursors
Two dendritic cell (DC) subsets have been identified in the murine system on the basis of their differential CD8a expression. CD8a 1 DCs and CD8a 2 DCs are considered as lymphoidand myeloidderived,
Splenic dendritic cell subsets prime and boost CD8 T cells and are involved in the generation of effector CD8 T cells.
TLDR
In vitro depletion of CD11c(+) DCs from mouse splenocytes significantly reduced the generation of pb9-specific IFNgamma producing effector CD8 T cells, indicating that splenic DCs are involved in the development of p b9- specific IFNGamma producingEffector cells.
A subset of human plasmacytoid dendritic cells expresses CD8α upon exposure to herpes simplex virus type 1
TLDR
The serendipitous finding that a subset of human PDC upregulates CD8α upon HSV-1 stimulation is reported, as shown by gene array and flow cytometry analyses and indicates a highly activated phenotype and appear to recruit other immune cells to the site of inflammation.

CD4 and CD8 expression by human and mouse thymic dendritic cells.
TLDR
The biological significance of CD4 and CD8 expression by DC is discussed in view of this major species difference and the possibility that human thymic DC may be targets for HIV infection.
Dendritic cell subtypes in mouse lymphoid organs: cross-correlation of surface markers, changes with incubation, and differences among thymus, spleen, and lymph nodes.
TLDR
Some aspects of this complex, but discrete, pattern of surface marker expression can be correlated with differences in lineage origin and functional activity of the DC.
Are CD8+ dendritic cells (DC) veto cells? The role of CD8 on DC in DC development and in the regulation of CD4 and CD8 T cell responses.
TLDR
The CD8+ DEC-205+ DC therefore differ in that respect from antigen-presenting 'veto' cells, where CD8 itself is involved in transmitting negative signals to the T cells, andDEC-205 may prove to be a more pertinent marker of the regulatory DC population.
A subclass of dendritic cells regulates the response of naive CD8 T cells by limiting their IL-2 production.
TLDR
Although CD8+ DC are very efficient at stimulating CD8 T cells into cell division, they are deficient at stimulating endogenous cytokine production, which means that the implications of these different DC regulatory systems are discussed.
Dendritic cell heterogeneity in vivo: two functionally different dendritic cell populations in rat intestinal lymph can be distinguished by CD4 expression.
TLDR
Two separate DC populations that differ functionally and phenotypically migrate from intestine to mesenteric nodes are found, which may reflect distinct DC lineages or differentiation modulated by different microenvironmental stimuli.
The nature of the signals regulating CD8 T cell proliferative responses to CD8α+ or CD8α− dendritic cells
TLDR
The CD8α−‐expressing dendritic cells of mouse spleen have been shown to be poor inducers of interleukin (IL)‐2 production by CD8 T cells when compared to the CD8− DC, and the difference in T cell proliferative responses was a consequence of differences in induced cytokine production.
A subclass of dendritic cells kills CD4 T cells via Fas/Fas-ligand- induced apoptosis
TLDR
FasL+ CD8+ DC are a specialized subgroup of DC with a role in the regulation of the response of primary peripheral T cells, and are shown to express FasL at high levels by direct surface staining.
CD8α+ and CD8α− Subclasses of Dendritic Cells Direct the Development of Distinct T Helper Cells In Vivo
TLDR
It is reported here that both subclasses of DCs efficiently prime antigen-specific T cells in vivo, and direct the development of distinct T helper (Th) populations, and it is shown that interleukin 12 plays a critical role in Th1 development by CD8α+ DCs.
Developmental pathways of dendritic cells in vivo: distinct function, phenotype, and localization of dendritic cell subsets in FLT3 ligand-treated mice.
TLDR
It is demonstrated that the mature DC subsets (C, D, and E) from Flt3 ligand-treated mice differ with respect to phenotype, geographic localization, and function, which underscores the importance of viewing DC development in vivo, as an interplay between distinct lineages and a maturational dependence on specific microenvironmental signals.
Assessment of CD4 expression by early T precursor cells and by dendritic cells in the human thymus.
TLDR
This work searched for a corresponding population of early T precursors in the infant human thymus, by first depleting the majority of more mature thymocytes, then using immunofluorescence and flow cytometry to analyze cells bearing a range ofEarly T lineage markers.
