Butanol (C4H10OH) has been used both to dissect the molecular targets of alcohols/general anesthetics and to implicate phospholipase D (PLD) signaling in a variety of cellular functions including neurotransmitter and hormone exocytosis. Like other primary alcohols, 1-butanol is a substrate for PLD and thereby disrupts formation of the intracellular signaling lipid phosphatidic acid. Because secondary and tertiary butanols do not undergo this transphosphatidylation, they have been used as… Expand
The Role of Phospholipase D in Regulated Exocytosis*
TLDR
The effects of phospholipase D activity and its product phosphatidic acid on the Ca2+ sensitivity and rate of fusion correlate with modulatory upstream roles in docking and priming rather than to direct effects on fusion per se. Expand
“Slow” Voltage-Dependent Inactivation of CaV2.2 Calcium Channels Is Modulated by the PKC Activator Phorbol 12-Myristate 13-Acetate (PMA)
TLDR
It is postulate that the kinetics of recovery from “slow” inactivation could provide a molecular memory of recent cellular activity and help control CaV2 channel availability, electrical excitability, and neurotransmission in the seconds-to-minutes timeframe. Expand
Jedi-1 deficiency increases sensory neuron excitability through a non-cell autonomous mechanism
TLDR
It is indicated that loss of Jedi-1 alters DRG neuron activity indirectly through an intercellular interaction between non-neuronal cells and sensory neurons, as well as alterations in the properties of voltage-gated sodium channel currents in Jedi- 1 null neurons. Expand
Membrane fluidization by alcohols inhibits DesK-DesR signalling in Bacillus subtilis.
TLDR
This work was able to suppress Des synthesis after a temperature downshift using linear alcohols and benzyl alcohol and found that the membrane fluidization induced by alcohols was reflected in an increased hydration at the lipid-water interface, associated with a decrease in DesK activity. Expand

Calcium signaling and exocytosis in adrenal chromaffin cells.
TLDR
It is proposed that chromaffin cells have developed functional triads that are formed by calcium channels, the ER, and the mitochondria and locally control the [Ca2+]c that regulate the early and late steps of exocytosis. Expand
5-Fluoro-2-indolyl des-chlorohalopemide (FIPI), a Phospholipase D Pharmacological Inhibitor That Alters Cell Spreading and Inhibits Chemotaxis
TLDR
5-fluoro-2-indolyl des-chlorohalopemide (FIPI) is examined, and it is shown that it rapidly blocks in vivo PA production with subnanomolar potency, indicating potential utility for it as a therapeutic for autoimmunity and cancer metastasis. Expand
Phospholipase activation and secretion: evidence that PLA2, PLC, and PLD are not essential to exocytosis.

TLDR
This is the first report of a Ca(2+)-independent PLA2 activity in human platelets, use of quercetin as a PLD inhibitor, and dissociation of PLA2, PLC, and PLD activities from secretion. Expand
Alcohols Inhibit a Cloned Potassium Channel at a Discrete Saturable Site
TLDR
The results strongly suggest that a discrete site within the ion channel protein is the primary locus of alcohol and general anesthetic action. Expand
ATP Serves as a Negative Feedback Inhibitor of Voltage-Gated Ca2+ Channel Currents in Cultured Bovine Adrenal Chromaffin Cells
TLDR
It is shown that stimulation of chromaffin cells results in the release of an "endogenous inhibitor" that suppresses I(Ca), and a negative feedback pathway is identified that may serve as an important regulatory mechanism for catecholamine secretion in chromaffIn cells. Expand
Gabapentin Inhibits Catecholamine Release from Adrenal Chromaffin Cells
TLDR
The work demonstrates that chromaffin cells are a useful model for additional investigation of the cellular mechanism(s) by which gabapentin controls neurosecretion and identifies altered adrenal catecholamine release as a potential contributor to some of the beneficial perioperative effects of gABapentin. Control of CaV2 Calcium Channels and Neurosecretion by Heterotrimeric G Protein Coupled Receptors
CaV2.1 (P/Q-type) and CaV2.2 (N-type) voltage-gated Ca2+ channels play pivotal roles in synaptic transmission and neuroendocrine hormone secretion by coupling excitation (i.e. action potentialExpand

TLDR
The mechanisms by which GPCRs inhibit Ca2+ channels in chromaffin cells are reviewed and how this might be altered by cellular context are discussed. Expand
Lack of Phospholipase D Activity in Chromaffin Cells: Bradykinin‐Stimulated Phosphatidic Acid Formation Involves Phospholipase C in Chromaffin Cells but Phospholipase D in PC12 Cells
TLDR
The results show that, although both neuronal models synthesize PA in response to bradykinin, they do so by quite different routes: PLC/DAG kinase for chromaffin cells and PLD for PC12 cells, which may indicate an essential role of PA in the regulation of cellular events by brady Kinin. Expand
Effect of an homologous series of aliphatic alcohols on neuronal and smooth muscle voltage-dependent Ca2+ channels.
TLDR
In synaptosomes, K(+)-stimulated 45Ca2+ uptake was 5-30 times more sensitive to the inhibitory actions of alcohol than were [3H]nitrendipine and [125I]omega-conotoxin binding, and this observation is supported by the observation that K( +)-stimulation 45Ca 2+ uptake which is insensitive to L- and N-channel antagonists was inhibited by funnel web spider venom.
