We report the use of bulked segregant SSR analysis for rapid identification of DNA markers linked to the Fr1 locus in soybean. Pooled DNA extracts from 10 homozygous Fr1Fr1 and 10 fr1fr1 F 2 plants, derived from a msMOS 3 Minsoy cross, were analyzed using 65 SSR markers. Five SSRs produced repeatable polymorphisms between paired bulks. Linkage with the Fr1 locus was tested using these five SSR primers and DNA from individual plants of each bulk. DNA polymorphisms generated by these five primers…

Excess heterozygosity contributes to genetic map expansion in pea recombinant inbred populations.

This analysis compares F(2) and RI maps derived from a cross between the same two parental lines and shows that excess heterozygosity contributes to map inflation.

Genetics and cytology of a genic male-sterile, female-sterile mutant from a transposon-containing soybean population.

The w4-m sterile was designated st8st8 and assigned Soybean Genetic Type Collection number T352 and Chromosome pairing, as observed in squash preparation, was almost completely absent in sterile plants.

2 Botany and Cytogenetics of Soybean

The cytogenetic knowledge of soybean lags far behind that of other model economically important crops because its somatic chromosomes are symmetrical and only one pair of satellite chromosomes can be identified.

Broadening the Genetic Base of Soybean: A Multidisciplinary Approach

The cytogenetic knowledge of soybean lags far behind that of other model important crops (rice, maize, wheat, tomato), because it's somatic chromosomes are symmetrical, and only one pair of satellite chromosomes can be identified.

Root fluorescent compounds from soybean controlled by the Fr loci

Soybean root fluorescence in ultraviolet light is controlled by the Fr loci and three closely related fluorescent compounds that are greatly reduced in extracts from non-fluorescent roots are found.

Microsporogenesis of Rps8/rps8 heterozygous soybean lines

A high frequency of meiotic abnormalities including fragments, laggards, multinucleated microspores; and microcytes containing DNA was also observed in Rps8/rps8 Williams × PI 399073 BC4F2:3 RILs, which may contribute to the high degree of segregation distortion present in the Williams-×-PI 399 073 populations.

Integration of Simple Sequence Repeat DNA Markers into a Soybean Linkage Map

Results indicated a relatively limited amount of clustering of soybean SSR loci, and demonstrated that microsatellite genetic markers should provide an excellent complement to RFLP and RAPD markers for use in soybean molecular biology, genetics, and breeding research.

An Integrated Genetic Linkage Map of the Soybean Genome

All but one of the classical linkage groups can now be assigned to a corresponding molecular linkage group on the basis of in situ segregation or linkage reports in the literature.

Brief communication. Assignment of the Fr3 locus to soybean linkage group 9

The gene order of Fr3-Ap-Ti-Lap1 (leucine aminopeptidase) was established for classical linkage group 9 in soybean and the linkage relationship between the loci Fr3 and Ap was ascertained.

Integration of the soybean molecular and classical genetic linkage groups

About half of the 19 soybean CLGs can now be associated with a corresponding MLG, as a result of the CLG to MLG assignments made in this study.

Chromosomal regions associated with segregation distortion of molecular markers in F2 , backcross, doubled haploid, and recombinant inbred populations in rice (Oryza sativa L.)

Ch Chromosomal regions associated with marker segregation distortion in rice were compared based on six molecular linkage maps, suggesting that loci in these regions may be associated with preferential regeneration of japonica genotypes during anther culture.

Identification of markers linked to disease-resistance genes by bulked segregant analysis: a rapid method to detect markers in specific genomic regions by using segregating populations.

Bulk segregant analysis has several advantages over the use of near-isogenic lines to identify markers in specific regions of the genome and will have widespread application both in those species where selfing is possible and in those that are obligatorily outbreeding.

Isolation of molecular markers from specific chromosomal intervals using DNA pools from existing mapping populations.

Although this method has been demonstrated in tomato, it should be applicable to any sexually reproducing organism for which segregating populations are being used to construct genetic linkage maps.

Use of isogenic lines and simultaneous probing to identify DNA markers tightly linked to the tm-2a gene in tomato.

The strategy outlined in this paper should be widely applicable for selecting DNA clones tightly linked to genes of interest, due to the availability of isogenic lines in many plant species.

Molecular mapping of segregation distortion loci in Aegilops tauschii.

Three regions of segregation distortion identified on chromosome 5D were analyzed in two sets of reciprocal backcross populations to analyze the effect of sex and cytoplasm on segregation distortion and agree with other studies stating that loci affecting gametophyte competition in male gametes are located on 5DL.
